采用析因设计优选溶解纤维连接蛋白配方的研究

摘  要:目的采用析因设计研究从几种化学试剂中筛选溶解纤维连接蛋白效果较优的试剂组合,观察该溶液是否对蛋白凝块有无容解作用,为清洗剂国产化做前期准备工作。方法利用两因素两水平析因设计的原理和方法,分别探讨尿素与吐温-80、氧化磷酸三钠与吐温-80溶解纤维蛋白凝块的效果,并与全自动血液凝固分析装置清洗液做对照,筛选出较优组合。结果氧化磷酸三钠与吐温-80组合及尿素与吐温-80的溶解血浆纤维蛋白凝块的效果与原装全自动血液凝固分析装置清洗液比较差异无显著性。结论筛选出的较优的溶纤维连接蛋白配方组合为氮化磷酸三钠和吐温-80,为替代原装清洗液运用于全自动血凝仪管道的清洁与维护做了初步探索。Objective To Select the formula that dissolve fibronectin preferably from several chemistry reagents is to be Observation of the solution is to have no protein clot dissolution, As a cleaning agent localization to do the preparatory work.Methods Explore the effect of Urea with Tween-80, Chlorinated sodium phosphate with Tween-80 to dissolve fibrin clot respectively by the principles and methods of two factors, two level of Factorial Design .At the same time, automatic blood coagulation analysis device cleaning solution is compared.Select the better formula from them. Results Phosphoric acid three sodium chloride and Twain -8 and Twain -80 of urea and the dissolution of plasma fibrin clot results with the original full automatic blood coagulation analyzer cleaning solution had no significant difference. Conclusion The screening out better formula to dissolve fibronectin is the combination of Chlorinated sodium phosphate with Tween-80 to replace the original cleaning fluid used in the automatic blood coagulation analyzer pipeline cleaning and maintenance
