摘 要:目的:探讨蜂胶总黄酮(total flavonoids of propolis,TFP)对脑缺血再灌注大鼠脑组织含水量及脑组织中一氧化氮(nitric oxide,NO)含量、一氧化氮合酶(nitric oxide synthase,NOS)表达的影响。方法:采用四血管阻断手术法建立大鼠全脑缺血再灌注模型,观察低、中、高剂量(25mg/kg、50mg/kg、100mg/kg)的TFP对全脑缺血再灌注后大鼠脑组织含水量、NO含量和NOS表达的影响。结果:与假手术组比较,模型组和各TFP组大鼠脑组织含水量、NO含量和NOS表达量均增高(P<0.01);与模型组比较,TFP 50mg/kg组和TFP 100mg/kg组大鼠脑组织含水量、NO含量、iNOS表达量均降低(P<0.01);与模型组比较,各TFP组大鼠脑组织TNOS活性均降低(P<0.01)。结论:TFP能明显降低脑缺血再灌注后脑组织含水量,抑制大鼠缺血再灌注后的脑组织中NO和NOS的表达。Objective:To investigate the effect of total flavonoids of propolis(TFP)on the water content,the content of nitric oxide(NO)and the expression of nitric oxide synthase(NOS)in cerebral ischemia-reperfusion brain tissue in rats.Methods:The model of global cerebral ischemia reperfusion injury was operated by pullsinelli-4vessel occlusion with SD rats of clean grade.The water content,the content of NO and the expression of NOS in cerebral ischemic area were measured in TFP groups of low,middle and high dose(25mg/kg,50mg/kg,100mg/kg).Results:Compared with the sham operation group,the water content,the content of NO and the expression of NOS were significantly increased in brain tissue in rats of the model group and three TFP groups(P〈0.01).Compared with the model group,the water content,the content of NO and the expression of iNOS in the brain tissue of group TFP 25mg/kg and group TFP 100mg/kg were significantly decreased(P〈0.01),and the activity of TNOS was significantly decreased in the three TFP groups(P〈0.01).Conclusion:TFP could obviously decrease the water content of brain tissue,inhibit the expression of NO and NOS in cerebral ischemia-reperfusion rats.
