摘 要:铜藻(Sargassum horneri)是浙江沿海重要的褐藻之一,具有较高的生态价值。对铜藻遗传多样性进行分析,可以为其资源保护、人工繁育以及海洋生态修复等提供理论参考。本研究利用AFLP分子标记技术,从64对选择性扩增引物中筛选出20对扩增效果好、带型清晰、条带分布均匀的引物,并用这20对引物对浙江4个铜藻群体进行了分析。结果显示,80个铜藻个体中共检测到有效结合位点407个,其中多态性结合位点270个;铜藻4个群体整体水平上的多态位点百分率(PPL)为66.34%,4个群体各自的多态位点百分率在30.49%~46.54%之间,均值为36.09%。4个铜藻群体的Nei基因多样性指数(H)为0.237 5,Shannon信息指数(I)为0.374 1,4个铜藻群体的总遗传变异(Ht)和群体内遗传变异(Hs)分别为0.314 2和0.256 9,群体间遗传分化系数(Gst)为0.182 3,即群体间遗传变异占总变异的18.23%,即群体内遗传变异占总变异的81.77%,表明群体间遗传分化程度较低。4个铜藻群体间的基因流动系数(Nm)为1.121 4,表明浙江沿海铜藻群体间有一定的基因交流。Sargassum horneri is one of the important brown algae in coastal waters of Zhejiang Province with high ecological value. Analysis into the genetic diversity of Sargassum horneri populations can provide theoretical reference for resource protection,artificial breeding,and ecological restoration,etc. In this study,the genetic diversity of 80 individuals from 4 populations of Sargassum horneri was analyzed by amplified fragment length polymorphism( AFLP)method. Twenty primer pairs were selected from 64 primer pairs for the AFLP experiment. It was shown that 407 band loci were detected,of which 270 were polymorphic. The percentage of polymorphic loci( PPL) was 66. 34%.The percentage of polymorphic loci in each population varied from 30. 49% to 46. 54%,with an average of 36. 09%.Nei's genetic diversity( H) was 0. 237 5,and Shannon's information index( I) was 0. 374 1. The total genetic diversity for species( Ht) and the mean heterozygosity within populations( Hs) were 0. 314 2 and 0. 256 9,respectively.The total Gstwas 0. 182 3,indicating that the major genetic variation was within populations,which accounted for81. 77%,and the minor variation was among populations,which accounted for only 18. 23%. The gene flow within4 populations was 1. 121 4,which reflected a certain amount of gene flow within the 4 populations in Zhejiang Province.
