摘  要:目的研究绞股蓝多糖对环磷酰胺所致免疫低下模型小鼠脾脏免疫功能的调节作用。方法建立免疫抑制小鼠模型,随机分为正常组、模型组、香菇多糖组、绞股蓝多糖低、中、高剂量组。14d后处死各组小鼠,检测小鼠体重、脾指数、脾淋巴细胞转化刺激指数、脾细胞上清及血清IgG、IgM含量、血清IL-4水平和脾细胞CD4+/CD8+比值。结果与模型组比较,绞股蓝多糖高剂量组明显提高免疫抑制小鼠体重、脾指数、脾淋巴细胞转化刺激指数及脾细胞CD4+/CD8+比值(P<0.05),绞股蓝多糖中、高剂量组显著升高血清IL-4水平(P<0.01),绞股蓝多糖低,中剂量组明显提升血清及脾细胞培养上清中IgG含量(P<0.05),绞股蓝多糖中、高剂量组显著升高血清及脾细胞培养上清中IgM水平(P<0.01)。结论绞股蓝多糖可以缓解环磷酰胺对小鼠脾脏免疫功能的抑制效应。Objective To determine if Gynostemma pentaphyllum Polysaccharide can alleviate suppression of mice induced by cyclophosphamide and to preliminarily explore the underlying mechanism. Methods Immunosuppressed murine model were established in Kunming mice induced by cyclophosphamide. Then these mice were randomly divided into normal group,model group,Lentinan group,Gynostemma pentaphyllum Polysaccharide low,middle and high dose group. After fourteen days of treatment,serum,spleen and spleen cell supernatant were taken out. Body weight,spleen index,the proliferation activities of splenic lymphocyte,Ig G,Ig M in serum and spleen cell supernatant,IL- 4 in serum and CD4+/ CD8+in spleen cell were detected. Results Compared with model group,body weight,spleen index,the proliferation activities of splenic lymphocyte and spleen cell CD4+/ CD8+proportion in Gynostemma pentaphyllum Polysaccharide high dose group was improved greatly( P〈0. 05),serum IL- 4 level in middle and high dose Polysaccharide group was increased( P〈0. 01),Ig G and Ig M of serum or spleen cell supernatant was improved( P〈0. 01 or P〈0. 05) in Gynostemma pentaphyllum Polysaccharide. Conclusion Gynostemma pentaphyllum Polysaccharide can alleviate cyclophosphamide- induced suppression of spleen immune function in mice.
