摘 要:目的优化中药东北天南星生物碱类成分提取、纯化工艺,确定最佳工艺参数。方法采用单因素考察结合正交实验设计,以胡芦巴碱含量、出膏率为指标筛选东北天南星生物碱类成分最佳提取工艺;采用大孔吸附树脂技术,通过静、动态吸附与解吸试验,优选最佳纯化工艺。结果中药东北天南星生物碱类成分最佳提取工艺为20倍量60%乙醇,回流提取2次,每次2h;最佳纯化工艺为HPD-300树脂,0.1g/ml浓度的提取液1∶1(药材∶干树脂)比例上样,5BV水洗脱除杂后,5BV 70%乙醇洗脱,以胡芦巴碱计,得到的天南星生物碱纯度提高了7倍。结论研究优化了东北天南星生物碱类成分的提取、纯化工艺,确定了最佳工艺参数,为其临床应用开发与工业化生产提供依据。Objective To optimize the extraction and purification process of alkaloids components in Arisaema amurense Maxim.and confirm the optimum process parameters. Methods Using single factor investigation combined with orthogonal experiment design,taking the content of trigonelline and extract rate as indexes to screen the optimal extraction process of alkaloids components;Using macroporous adsorption resin technology,through static and dynamic adsorption and desorption experiment,to optimize the best purification process. Results The optimal extraction process is 20 times 60% ethanol,circumfluence extraction 2 times,each time 2 h; the best purification process is HPD- 300 type resin,0. 1 g / m L extracting solution on 1: 1 proportion( herbs: dry resin),5 times the amount of cylinder water removing impurity,5 times 70% ethanol elution,the purity of alkaloids increased 7times for trigonelline is calculated. Conclusion In this study,we optimized the extraction and purification process of alkaloids components in Arisaema amurense Maxim.,determined the optimum process parameters,which provide a basis for its clinical application and industrial production.
