不同壮数艾炷灸神阙穴对衰老豚鼠SOD蛋白表达及基因表达的影响

摘　　要：目的观察不同壮数艾炷灸神阙穴对衰老豚鼠超氧化物歧化酶(Superoxide dismutase,SOD)蛋白表达及相关基因表达的影响。方法采用D-半乳糖腹腔注射建立豚鼠亚急性衰老模型,用酶联免疫吸附试验(Elisa)和免疫组化染色观察各组SOD蛋白表达,用实时荧光绝对定量聚合酶链反应(Realtime Polymerase Chain Reaction,RT-PCR)法检测豚鼠肾组织相关基因表达。结果造模后除正常组外,其余各组豚鼠血清SOD含量均较造模前明显降低(P<0.05),提示造模成功。各治疗组血清SOD含量、基因表达量明显高于模型组(P<0.05),与正常组相比无差异(P>0.05);各治疗组组间比较,疗效无差异(P>0.05)。免疫组化染色示:各治疗组切片阳性率与正常组无差异(P>0.05),明显高于模型组(P<0.05);各治疗组阳性率组间比较无差异(P>0.05)。结论艾炷灸神阙穴可提高衰老豚鼠血清SOD含量及其相关基因表达,但不同剂量的艾炷灸对其影响在本实验中未见明显差异。通过免疫组化染色发现艾炷灸神阙穴可提高SOD在衰老豚鼠肾组织中的蛋白表达。Objective To observe the effect of different doses of moxa cone moxibustion Shenque on the expression of SOD（ Superoxide dismutase） protein and related genes in senescent guinea pigs. Methods Established the aging model by intraperitoneal injection of D- galactose in guinea pigs. The expression of SOD protein was observed by Elisa and immunohistochemical staining.The expression of related genes in renal tissue of guinea pigs was detected by the PT- PCR（ Realtime Polymerase Chain Reaction）. Results After modeling,except the normal group,the serum SOD levels of other groups in guinea pig were significantly decreased than before making model（ P〈0. 05）,suggesting the model is successful. The serum SOD content,gene expression in treatment groups was significantly higher than model group（ P〈0. 05）,while no difference compared with the normal group（ P〈0. 05）; there was no difference on curative effect among the treatment groups（ P〈0. 05）. Immunohistochemical staining effect showed： there was no difference on the positive rate of biopsy between the treatment groups and the normal group（ P〈0. 05）; the positive rate of biopsy in the treatment groups was significantly higher than that of the model group（ P〈0. 05）; there was no difference on positive rate among the treatment groups（ P〈0. 05）. Conclusion Moxa cone moxibustion can increase the expression of serum SOD content and the related gene in senescence guinea pigs,but there was no significant difference on the effects of different doses of moxa cone moxibustion in this experiment. Moxa cone moxibustion on Shenque can increase expression of SOD protein in the renal tissue of senescent guinea pigs from immunohistochemical staining.
