摘 要:目的探索肝癌中特异性表达的microRNA与所调控的靶基因网络关系、差异基因与细胞通路改变的关系及不同证型组间的表达差异。方法通过对肝癌组织样本进行miRNA芯片技术进行筛选,获得不同证型肝癌组织与正常肝组织中差异性表达miRNA,再用GO分析对这些miRNA进行靶基因预测和靶基因功能富集分析,得到网络调控的关键miRNA和受调控作用的关键靶基因及其功能,最后用PCR验证相关miRNA及比较不同中医证型组表达的差异。结果初步筛选出肝癌组织中明显上调的关键miRNA12个,下调的关键miRNA10个,GO分析发现靶基因与调控其他基因表达、细胞分化、生长、增殖、凋亡、生长因子受体通路、细胞粘附、血管生成和细胞运动等多项生物学过程有关。进一步的PCR验证也发现不同证型组间存在miRNA表达的差异。结论肝癌组织中差异性表达的miRNA及其所调控的靶基因与肝细胞发生癌变、癌细胞转移和肿瘤内血管生成等多个细胞学进程中的多个靶点起到关键作用,不同证型组肝癌组miRNA表达谱具有独自的特征。Objective To find the network of hepatocellular carcinoma specific microRNAs and their target genes. Methods The liver tissue samples from surgery resection were analyzed by miRNA array to obtain the differently expressed miRNAs between liver cancer tissue and normal tissue,then the GO analysis was used to identify the target genes. Results 12 up- regulated and 10down- regulated key miRNAs in liver cancer were found after primary screening,GO analysis demonstrated that these target genes were associated with regulation of gene expression,cell differentiation,development,proliferation,apoptosis,adhesion,motion,and angiogenesis,growth factor receptor pathway. Conclusion The liver cancer specific miRNAs and their target genes play crucial roles in cell process of liver tumorgenesis including hepatocytes cancerization,metastasis and angiogenesis in tumor. In addition,different TCM syndromes have their characteristic miRNA profiles
