通腑泄热法对脑膜炎发热大鼠神经肽类分子的影响

摘　　要：目的基于脑-肠轴学说,探讨通腑泄热法对脑膜炎发热大鼠神经肽类分子的调节作用。方法采用小脑延髓池接种埃希氏大肠杆菌混悬液建立大鼠细菌性脑膜炎发热模型。实验动物随机分为6组:空白组、模型组、大承气汤组,醒脑静注射液组、大承气汤合醒脑静注射液组、扑热息痛组。检测各组动物用药后脑、脊髓、结肠、肺、胃、肾组织、血浆中CRH、AVP、P物质、NT水平的变化。结果与模型组相比,通腑泄热组中脑肠等组织及血浆中CRH含量降低、AVP含量升高、P物质含量降低、NT含量升高,有显著性差异(P<0.05),而与其他组相比,变化无显著差异性。结论通过脑-肠轴来调节神经肽类介质的释放,脑肠互动,可能是通腑泄热法治疗脑病发热的机制。Objective To investigate regulation of neuropeptides of Tongfu Xiere method for meningitis fever in rat,based on the theory of brain- gut axis. Methods The rat fever model induced bacterial meninggitis was established with injecting the Escherichia coli into Cerebellomedullary cistern. The all experimental animals divided into six groups： the normal group,the model group,Large Chengqi Decoction group,Xingnaojing Injection group,Large Chengqi Decoction and Xingnaojing Injection group,Paracetamol group. The levels of corticotropin releasing hormone（ CRH）,arginine vasopressin（ AVP）,substance P（ SP） and neurotensin（ NT） were measured by enzyme- linked immunosorbent assay in hypothalamus,spinal cord,colon,lung,stomach,kidney,and blood after each group treated by respectively different drugs. Results Compared with the model group,the levels of CRH and Sp in brain,bowels,and etc. were significantly reduced,but the lever of AVP and NT were significantly elevated in the Tongfu Xiere group（ P〈0. 05）. Compared with the other groups,these differences were not funded. Conclusion Tongfu Xiere method can adjust the levels of neuropeptides in brain and bowels,which may be mediated by brain- gut axis. It is the mechanism of Tongfu Xiere therapy for fever induced by encephalopathy.
