Effects of antioxidants on the ageing and physiological indexes of Phalaenopsis protocorm-like bodies in long-term subculture

摘  要:为减缓长期继代蝴蝶兰类原球茎增殖、萌发能力降低,在培养基中添加抗氧化剂继代类原球茎24个月,观测类原球茎增殖能力、成活率及萌发率,并分析其生理学指标。结果表明:100 mg/L半胱氨酸与8.0 g/L甘露醇组合为适宜抗氧化剂浓度;在培养第24个月,处理组每瓶鲜重较对照组显著提高,萌发率极显著提高;培养期间对照组和处理组类原球茎的总抗氧化能力、超氧化物歧化酶、过氧化物酶活性均有不同程度的下降,处理组总抗氧化能力、超氧化物歧化酶、过氧化氢酶活性较对照极显著提高(主要在第16个月),对照组与处理组叶绿素水平均变化不明显。To slow down the decline of proliferation and germination capabilities of Phalaenopsis protocormlike bodies(PLBs)in long-term subculture,antioxidants were added to the media,the PLBs' proliferation capability and survival and germination rates were observed in 24-month subculture,and their physiological indexes were also analyzed.The results showed that a suitable antioxidant combination was 100 mg/Lcysteine and 8.0 g/L mannitol;The treatment group' s fresh weight per culture bottle in the 24 th month was significantly greater than that of the control,and the PLBs' germination rate was improved very significantly;During PLBs subculture all the total antioxidant capacity and SOD and POD activities decreased to different extents in both treatment group and control group,but all the total antioxidant capacity and SOD and CAT activities in the treatment group were very significantly higher than those in the control group(mainly in the 16 th month),yet their chlorophyll contents in both groups did not very obviously.
