摘 要:目的 研究胸腺素β4对人增生性瘢痕成纤维细胞中基质金属蛋白酶-2(MMP-2)和MMP-9的表达影响,探讨胸腺素β4对增生性瘢痕的作用机制。方法 2013年6-9月在南昌大学第一附属医院烧伤整形科手术切取的增生性瘢痕组织(烧伤后6个月内),体外培养成纤维细胞,分为实验组(胸腺素浓度分别为0.05、0.1、1.0、5.0μg/mL),对照组(不添加胸腺素β4),采用反转录-聚合酶链反应(RT-PCR)以及蛋白免疫印迹法(Western blot)测定实验组和对照组的MMP-2、MMP-9表达变化。结果胸腺素β4作用后增生性瘢痕组织内MMP-2、MMP-9水平增加,分别以1.0μg/mL实验组和5.0μg/mL实验组作用最明显,与对照组比较差异均有统计学意义(P〈0.05)。结论 胸腺素β4能剂量依赖性促进MMP-2、MMP-9表达,可能是抑制增生性瘢痕的作用因素之一。Objective To study the effect of thymosin beta4 on the expression of matrix metalloproteinase-2(MMP-2) and MMP-9 of human hypertrophic and explore mechanism of thymosin beta4 on hypertrophic scar. Methods Fibroblasts were cultured in vitro ic patients(within 6 months after burns) admitted in the first affiliated hospital of Nanchang university from Ju: 2013. The Fibroblasts were divided into experiment group:0.05,0. 1,1, 0μg/mL and 5.0 μg/mL,and control grou eta4. The expression levels of MMP-2-,MMP-9 were observed by RT-PCR and Western blot. Results The expression MMP-2 and MMP-9 were increased after the thymosin beta4 took effect , and it was especially significant in 1. 0 μg/mL and 5. 0μg/mL group,there was statistical difference between experlment group and control group (P〈0. 05). Concision Thymosin beta4 can dose-dependently promote the expression of MMP-2 and MMP-9 and inhibit the pro-liferation of hypertrophic scar.
