摘  要:目的:观察采用微量注射泵经侧脑室给予中药芪苈强心胶囊对慢性心衰大鼠血流动力学、神经内分泌因子的影响。方法:采用结扎冠状动脉左前降支建立急性心肌梗死致大鼠心衰模型,造模4周后经侧脑室分别给予芪苈强心脑脊液和芪苈强心溶液,以阳性药CRH竞争性抑制剂(αh-CRH)作为对照,给药4周后应用颈动脉插管测定各组大鼠血流动力学指标,ELISA法检测大鼠血清脑利钠肽(BNP)、血浆去甲肾上腺素(NE),RT-PCR法检测下丘脑促肾上腺皮质激素释放激素(CRH)mRNA与脑垂体促肾上腺皮质激素(ACTH)mRNA的表达。结果:芪苈强心脑脊液组、芪苈强心溶液组、阳性药组均可改善心衰大鼠血流动力学,降低大鼠血清BNP、血浆NE,并可降低下丘脑组织CRH mRNA与脑垂体组织ACTH mRNA的表达。结论:经侧脑室给予复方中药芪苈强心可有效改善大鼠血流动力学,降低血液中神经内分泌因子的含量与下丘脑CRH mRNA、脑垂体ACTH mRNA的表达,影响心衰大鼠中枢改变,有助于治疗心衰。Objective: To investigate the impact of the lateral ventricle administered by syringe pump on blood flow dynamics and neuroendocrine factors in chronic heart failure rats. Methods: To establish the acute myocardial infarctioninduced heart failure in rats by the left anterior descending coronary artery ligation. The rats were given Qiliqiangxin cerebrospinal fluid,Qiliqiangxin solution and CRH competitive inhibitor( αh-CRH) by the left ventricle for four weeks after modeling. Four weeks after administration,carotid artery was used to measure blood flow dynamics indicators in rats in each group,ELISA was used to detect serum brain natriuretic peptide( BNP) and norepinephrine( NE),and PCR was used to detect the expression of the hypothalamus CRH mRNA and the pituitary ACTH mRNA. Results: Qiliqiangxin cerebrospinal fluid group,Qiliqiangxin solution group and positive drug group can improve blood flow dynamics,lower serum BNP,plasma NE,and reduce the expression of CRH mRNA and ACTH mRNA in rats with heart failure.Conclusion: Giving compound Chinese medicine by the lateral ventricles can improve hemodynamics,reduce blood levels of neuroendocrine factors,lower the expression of CRH mRNA and ACTH mRNA,contribute to the treatment of heart failure,and impact the change of the central nervous in heart failure rats.
