摘 要:目的:观察槲皮素对2型糖尿病大鼠肾脏肥大的影响并探讨其作用机制。方法:建立2型糖尿病大鼠模型,按随机数字表法分为正常对照组、糖尿病模型组以及槲皮素组,测定各组大鼠血糖、血脂、总胆固醇、血清胰岛素、胰岛素敏感指数(ISI)、肾脏肥大指数和单根近端小管(PT)钠钾ATP酶(Na+,K+-ATPase)活性,免疫组化法测定肾脏Toll样受体4(TLR4)蛋白表达。结果:与模型组比较,槲皮素组大鼠血糖、血脂、总胆固醇、血清胰岛素水平、肾脏肥大指数、Na+,K+-ATPase活性和肾内TLR4蛋白表达显著降低,差异均有统计学意义。结论:槲皮素能抑制2型糖尿病大鼠肾脏肥大,其机制可能与降低血糖、血脂、总胆固醇和肾脏TLR4蛋白表达,升高ISI及稳定Na+,K+-ATPase活性有关。Objective: To observe the effect of quercetin on diabetic rats ' kidney hypertrophy and explore its mechanism. Methods: The model of type 2 diabetic rates was made. The rats were randomly divided into three groups:normal group,diabetic model group,and quercetin group. The blood sugar,blood fat,total cholesterol,serum insulin level,insulin sensitivity index( ISI),KW / BW,Na+,K+-ATPase activity of PT were measured. The toll-like receptor 4( TLR4)protein expression was measured by immunohistochemical method. Results: Compared with the model group,The blood sugar,blood fat,total cholesterol,serum insulin level,KW / BW,Na+,K+-ATPase activity of PT,TLR4 protein expression were significantly reduced,so the differences were statistically significant. Conclusion: Quercetin can inhibit kidney hypertrophy,it may be caused by the decrease of blood sugar,blood fat,cholesterol,TLR4 protein expression and increase of ISI and stabilization of Na+,K+-ATPase activity of PT in T2 DM rats.
