针刀干预对膝骨关节炎兔软骨细胞整合素β1表达的长期影响

摘　　要：目的:探讨针刀对KOA的长期作用。方法:按随机数字表法将48只兔分为空白组、模型组、针刀组、电针组各12只,后3组用伸直位固定制动法造模,治疗后取材检测软骨整合素β1mRNA、蛋白。结果:模型组整合素mRNA水平降低,较空白组比较差异有统计学意义;针刀组整合素mRNA水平上升,较空白组比较差异无统计学意义;电针组整合素蛋白水平上升,较空白组差异显著。结论:针刀可上调整合素β1基因和蛋白水平,长期观察发现其含量接近正常,即针刀的长期疗效是显著的。Objective To explore the long-range mechanism of needle knife lysis on knee osteoarthritis. Methods： Sixmonth-old New Zealand rabbits were randomly divided into normal control group,model group,acupotomy group,and electro-acupuncture（ EA） group. KOA model was established by videman,After modeling,acupotomy group and electroacupuncture group were treated for three weeks,once every week in acupotomy group,and three times in electroacupuncture group. After the treatment,continued to normal breeding three weeks,then testted rabbits＇ behavior with knee level assessment method; Expression of apoptosis genes integrinβ1 mRNA,was conducted by Real-time PCR test,and expression of apoptosis proteins integrinβ1,was conducted by Western blot test. Results The results of Real-time PCR tests： after modeling,Integrin β1 mRNA expression levels in the rabbit knee cartilage extremely reduced; compared with normal control group showed an extreme difference. After acupotomology intervention and continued to normal breeding three weeks,compared with model group,Integrin β1 mRNA expression levels rose. There was no significant difference between the normal control group and the acupotomology group. Compared with normal control group,Integrin β1 mRNA expression level rose in electro-acupuncture,showing an extreme difference. The results of western blot tests： after modeling,integrinβ1 protein levels in the rabbit knee cartilage reduced abnormally,compared with normal control group showed a significant difference. Acupotomology intervention raised the integrin β1 protein expression levels,and electricity for this regulation raised the integrin β1 protein expression levels. Conclusion： Acupotomology not only can adjust the mechanical signals stimulate cartilage cells and raise the gene and protein expression levels of integrinβ1,but also can improve cartilage matrix. In the long-term observation its content closed to normal. This illustrates the long-term efficacy of Acupotomology is positive.
