摘 要:目的:使用线性离子阱傅立叶转换串联质谱仪,对丹参中丹酚酸A和B进行多级裂解分析,探析其裂解规律。方法:采用进样针直接进样的方法,使用碰撞诱导解离(CID)裂解模式,对丹酚酸A和B进行多级裂解分析,Xcalibar V2.0进行数据分析,对碎片离子分子式推导,偏差设置为20。对两个化合物的裂解进行初步分析,全扫描及多级碎裂扫描的分辨率为30 000,多级碎裂模式为CID,碎裂能为35%。结果:丹酚酸类化合物在多级碎裂中,主要表现为丹参素基的丢失,且CO和H2O的丢失也较为明显。结论:本研究使用LTQ-Orbitrap高分辨质谱对两个化合物裂解途径进行分析,探讨其可能的裂解规律,为快速对中药材中该化合物的鉴别及其代谢物的鉴别提供实验数据支撑和思路。Objective:In this study,LTQ-Orbitrap high-resolution MS was used to explore the main fragmentation of salvianolic acids A and B using the CID type,which were isolated from the roots and rhizomes of Salvia miltiorrhiza.Methods:The standard solution were injected into the MS to gain the MSn data of the two constituents,respectively,the resolution of the full scan and MSn scan was 300 00,the fragmentation type was CID,and the energy was 35%.Results:The salvianolic acids A and B were a kind of phenolic acid constituents,it had a good response under the negtive model ESI-MS.The fragmentation of salvianolic acids were mainly for the loss of danshensu group,and CO and H2O were also obvious.Conclusion:The LTQ-Orbitrap was used to reveal the fragmentation pathway of salvianolic acids A and B from which the MSn fragment ions were obtained,and it will provide a high-efficiency and sensitive method to identity the two constituents from the roots and rhizomes of S.miltiorrhiza and its related metabolites.
