摘  要:目的:探索和络泄浊颗粒对大鼠肾脏纤维化中miR-200a表达的影响及探讨其可能存在的作用机制。方法:30只SD雄性大鼠随机分为6组:假手术组(Sham)、手术组(UUO)及和络泄浊颗粒(UUO+REG)组各2组,运用单侧(左)输尿管结扎法制造肾间质纤维化模型,但Sham组仅游离输尿管,而其余两组则游离并结扎输尿管。术后各组按1 mg/kg·d-1的量进行灌胃,UUO+REG组给予REG,其余两组则予生理盐水。术后第7天和14天,摘除左梗阻侧肾脏进行免疫组化检查α-SMA和荧光定量PCR检测miR-200a的表达。结果:在UUO及UUO+REG组,α-SMA表达明显高于Sham组(P<0.01);UUO+REG组低于UUO组(P<0.05)。miR-200a表达水平在UUO组和UUO+REG组都是降低的,但是UUO组与Sham组差别明显(P<0.01),其与UUO+REG组亦有明显的差别(P<0.05)。结论:和络泄浊颗粒可以通过上调miR-200a延缓肾脏纤维化进展。Objective: To investigate the effects and mechanism of action of REG on micro RNA-200 a expression in renal interstitial fibrosis rat that was established by unilateral ureter obstruction. Methods: Thirty male Sprague-Dawley(SD) rats were randomly divided into six groups(5 mice/group): two sham operation groups(Sham), two model groups(UUO), and two drug groups(UUO+REG). Renal interstitial fibrosis was established in UUO and UUO+REG groups, but not in Sham groups, by unilateral ureter obstruction. The left Kidney was harvested from all groups either 7 d or 14 d post-operation and subsequently examined for changes inα-SMA or micro RNA-200 a expression. Results: The expression of α-SMA increased significantly(P〈0.01) in both UUO and UUO+REG groups at 7 d and 14 d post-operation; however, there was a significant difference in α-SMA expression in UUO+REG group compared with UUO group(P〈0.05). Finally, the expression levels of miR-200 a were reduced in both UUO and UUO+REG groups as compared with that in Sham groups at 7 d and 14 d post-operation, while the expression in UUO group was significantly lower than that in Sham(P〈0.01) and UUO+REG groups(0.05)at 7d and 14 d post-operation. Conclusion: REG could delay the progression of chronic renal failure by up-regulating miR-200 a.
