长春碱亲水基修饰阳离子脂质体对荷瘤小鼠的抗肿瘤作用研究

摘　　要：目的:研究长春碱亲水基修饰阳离子脂质体对荷瘤小鼠的抗肿瘤作用。方法:腋下接种S180腹水瘤小鼠黄色腹水复制荷瘤模型。将荷瘤小鼠随机分为模型组、硫酸长春碱注射液组、长春碱普通脂质体组、长春碱亲水基修饰脂质体组、长春碱阳离子脂质体组和长春碱亲水基修饰阳离子脂质体组,分别记为A、B、C、D、E、F组,每组18只。A组小鼠尾iv生理盐水,其余各组小鼠尾iv长春碱(给药剂量为1.5 mg/kg),每隔1日给药1次,连续给药5次。以小鼠生存状态、生存时间、瘤体积、瘤质量及组织病理切片为指标比较长春碱不同制剂的抗肿瘤作用。结果:与A、B、C、D、E组比较,F组小鼠更活跃、生存时间更长、瘤体积更小、瘤质量更轻,差异具有统计学意义(P<0.05);组织病理切片显示,F组小鼠肿瘤细胞核凝固坏死、核破碎溶解。结论:长春碱亲水基修饰阳离子脂质体有明显的抗肿瘤作用,且作用优于上述长春碱的其他制剂。OBJECTIVE：To study the anti-tumor effects of Vinblastine（VLB）hydrophilic group modified cationic liposomes in tumor-bearing mice. METHODS：Tumor-bearing model were induced by inoculating yellow ascites of S180 ascites tumor mice.Tumor-bearing mice were randomly divided into model group,VLB sulfate injection group,VLB liposomes group,VLB hydrophilic group modified liposomes group,VLB cationic liposomes group and VLB hydrophilic group modified cationic liposomes group,i.e. group A,B,C,D,E and F,with 18 mice in each group. Group A was given normal saline intravenously via mice tail,other groups were given VLB 1.5 mg/kg every 2 days for consecutive 5 times. The anti-tumor effects of different VLB preparations were compared,using living conditions,survival time,tumor volume and weight,and tissue pathological section as indexes. RESULTS：Compared with group A,B,C,D and E,the mice of group F were more active,and had longer survival time,smaller tumor volume and lighter tumor weight,with statistical significance（P〈0.05）. The tissue pathological section of mice in group F indicated that coagulation necrosis,disintegration,and dissolution of tumor cell nucleus. CONCLUSIONS：VLB hydrophilic group modified cationic liposomes have obvious anti-tumor effect,which are better than other VLB preparations.
