摘 要:目的:建立HPLC法同时测定氯柳酊中氯霉素和水杨酸的含量。方法:色谱柱为C18柱(4.6 mm×250 mm,5μm);流动相为甲醇-水-冰醋酸(50∶50∶1);流速为1.0 m L·min-1;柱温为30℃;检测波长为295 nm;进样量为10μL。结果:氯霉素和水杨酸分离良好。氯霉素在60.77-141.80μg·m L-1范围内线性关系良好(r=0.999 8),水杨酸在123.50-288.10μg·m L-1范围内线性关系良好(r=0.999 7),氯霉素和水杨酸的平均回收率分别为99.76%、99.87%,RSD分别为1.08%(n=9)、1.19%(n=9)。结论:本方法简便准确,快速可靠,可用于氯柳酊的含量测定和质量控制。Objective: To establish the HPLC method for simultaneous determination of chloramphenicol and salicylic acid in tincture. Methods: HPLC was used in the quantitative analysis with C18 chromatographic column(4.6 mm × 250 mm, 5 μm). The mobile phase consisted of methanol-water-glacial acetic acid(50 : 50 : 1) with the flow rate of 1.0 m L·min-1. The column temperature was 30 ℃. The detection wavelength was 295 nm and the injection volume was 10 μL. Results: The calibration curves of chloramphenicol and salicylic acid had a good linearity in the range of 60.77 – 141.80 μg·m L-1(r = 0.999 8) and 123.50 – 288.10 μg·m L-1(r = 0.999 7) respectively. The average recovery of chloramphenicol was 99.76% with RSD of 1.08%(n = 9). The average recovery of salicylic acid was 99.87% with RSD of 1.19%(n = 9). Conclusion: The method is simple, accurate and reliable, which can be used as the quality control method of this preparation.
