摘  要:目的评价单磷酸腺苷激活蛋白激酶(AMPK)激活剂对老年大鼠全脑缺血再灌注海马神经元凋亡的影响。方法老年雄性SD大鼠108只,随机分为假手术组(S组)、缺血再灌注组(I/R组)、5-氨基-4-咪唑甲酰胺核苷酸(AICAR)组(A组),每组36只。于再灌注12、24和48h时,每组随机取12只大鼠处死,取海马组织,采用TUNEL法测定细胞凋亡率,采用Western blotting法检测磷酸化AMPKα(p-AMPKα)的表达。结果与S组比较,I/R组和A组各时点海马神经元凋亡率升高;与I/R组比较,A组12、24h海马神经元凋亡率降低(F=5.12~16.54,P〈0.05、0.01)。I/R组和A组各时点海马神经元p-AMPKα水平明显高于S组,A组明显高于I/R组(F=6.30~18.29,P〈0.05、0.01)。结论 AMPK激活剂预处理可上调全脑缺血再灌注老年大鼠p-AMPKα表达水平,减少海马神经元凋亡。Objective To evaluate the effect of AMP-activated protein kinase activator on apoptosis in hippocampal cells induced by global cerebral ischemia-reperfusion(I/R)in aged rats. Methods This study consisted of 108 aged male SDrats,which were equally randomized to sham operation group,I/R group and AICAR pretreatment group(AICAR group),with 36 rats in each group.At 12,24 and 48hours of reperfusion,12 rats were sacrificed at random in each group,the hippocampus tissue was taken to detect its apoptotic rate of the cells using TUNEL method,and the expression of phosphorylated AMPKα(p-AMPKα)was detected by using Western blotting. Results Compared with sham operation group,neuronal apoptosis rate was significantly increased at each time point in I/R group and AICAR group;compared with I/R group,apoptosis rate significantly decreased at12 and 24hours in AICAR group(F=5.12-16.54;P〈0.05,0.01).The expression of p-AMPKαwas significantly higher,at each time point,in I/R group and AICAR group than that in sham operation group,and that in AICAR group was higher than that in I/R group(F=6.30-18.29;P〈0.05,0.01). Conclusion Pretreatment with AMP-activated protein kinase activator can upre-gulate the expression of p-AMPKαand decrease neuronal apoptosis induced by global cerebral ischemia-reperfusion in aged rats.
