摘  要:目的探讨气虚病理状态对皮肤吸收和氮酮促透功能的影响。方法将正常家兔皮肤分为2组,A组为药物组,B组为药物加1%氮酮组,气虚家兔皮肤分为4组,C组为气虚皮肤药物组,D组为药物加1%氮酮组,E组为药物加2%氮酮,F组为药物加4%氮酮组,采用双室透皮扩散装置进行茶碱体外经皮渗透实验,分别于2 h、4h、8 h、12 h和24 h取样,采用紫光光度计检测茶碱。计算累积渗透量(Q)、渗透系数(Kp)、稳态流量(Jss)、滞后时间(Tlag)和增渗倍数(ER),进行统计分析。结果 B组4 h、8 h、12 h和24 h累积渗透量均高于A组(t=5.65,P<0.01;t=7.21,P<0.01;t=8.66,P<0.01;t=7.23,P<0.01);C组2 h、4 h、8 h、12 h和24 h累积渗透量均高于A组(t=2.63,P<0.01;t=3.19,P<0.01;t=3.34,P<0.01;t=3.46,P<0.01;t=3.07,P<0.01);D组2 h、4h、8 h、12 h和24 h累积渗透量均高于C组(t=2.98,P<0.01;t=5.13,P<0.01;t=9.50,P<0.01;t=12.74,P<0.01;t=20.98,P<0.01),累积渗透量为E组>D组>F组>C组;C组和A组在JSS和Kp、Tlag差异均无统计学意义(P>0.05);D组、E组和F组Jss和Kp均高于C组(P<0.01)。结论气虚大鼠皮肤对药物的通透性增加,2%氮酮在气虚家兔皮肤的促透效果最佳。Objective To study the changes of skin absorption and azone transdermal function under Qixu pathological conditions. Methods Noimal rabbit skins were divided into two groups, A group ( drugs groups ), B group ( drugs + 1% azone), qixu rabbit skin were divided into four groups, Group C (qixu skin), group D (1% azone group ), E group (2% Azone) ,F group(4% azone) in vitro percutaneous permeation experiments with two-chamber transdermal diffusion. The samples were collected at 2 h, 4 h, 8 h, 12 h and 24 h, respectively. The concentration of theophylline was detected by UV. The cumulative permeation amount ( Q), permeability coefficient ( Kp ) , steady-state flow ( Jss ), lag time ( Tlag ) and enhancing rate(ER) were calculate and statistically analyed. Results In 2 h,4 h, 8 h, 12 h and 24 h the cumulative penetration, Group B were higher than group A ( t = 5.65, P 〈 0.01 ; t = 7.21, P 〈 0. 01 ; t = 8.66, P 〈 0. O1 ; t = 7.23, P 〈 0. 01 ), Group C were higher than group A ( t = 2.63, P 〈 0. 01 ; t = 3.19, P 〈 0. 01 ; t = 3.34, P 〈 0.01 ; t = 3.46, P 〈 0. 01 ;t = 3.07,P 〈0. 01 ), Group D were higher than group C ( t = 2.98, P 〈 0. 01 ; t = 5.13, P 〈 0. 01 ; t = 9.50, P 〈 0.01 ;t = 12.74,P 〈 0. 01 ; t = 20.98, P 〈 0. 01 ). The sequence of cumulative permeation were E group 〉 D group 〉 F group 〉 C group. Group C and Group A in Jss and Kp ,There was no significant difference ( P 〉 0.05 ). In Jss and Kp, D group, E group and F group were higher than group C (P 〈 0. 01 ). Conclusion Under qixu condition, the skin penetration was increased,2% Azone has a best skin penetration effect.
