摘  要:目的研究海马CA1区微量注射ghrelin对C57Bl6J小鼠海马依赖的记忆获取的影响。方法小鼠背侧海马CA1区微量注射5μmol/L的ghrelin 0.5μL,30min后进行旷场实验、高架十字迷宫实验及Morris水迷宫、条件恐惧测试。结果与对照组相比,背侧海马CA1区微量注射ghrelin可抑制小鼠空间记忆和场景恐惧记忆的获取(t=2.8~7.1,t′=9.8,P〈0.05)。与对照组相比,CA1区微量注射ghrelin不影响小鼠的自发活动和焦虑状态。结论背侧海马CA1区微量注射ghrelin对海马依赖的记忆获取产生抑制作用。Objective To study the effect of micro-infusion of ghrelin into CA1 on hippocampus-dependent memory acquisition in C57Bl6 Jmice. Methods Ghrelin(5μmol/L,0.5μL/per side)was micro-infused into CA1 region of the dorsal hippocampus.Behavioral training or testing was conducted 30 min after injection.The behavioral tasks applied were open field,elevated plus maze,Morris water maze and contextual fear condition. Results Compared with the normal saline control group,micro-infusion of ghrelin(5μmol/L,0.5μL/per side)into CA1 during training blocked both spatial memory formation and contextual fear memory acquisition as tested in Morris water maze and fear conditioning(t=2.8-7.1,t′=9.8,P〈0.05).Ghrelin treatment did not change locomotion activity and anxiety of those animals. Conclusion Micro-infusion of ghrelin into CA1 may suppress hippocampus-dependent memory.
