ASSOCIATION OF IL-8-251 GENE POLYMORPHISM WITH THE SUSCEPTIBILITY OF ENTEROVIRUS-71 ENCEPHALITIS

摘  要:目的探讨白细胞介素8(IL-8)-251位点基因多态性对肠道病毒71型(EV71)脑炎患病风险及病情程度的影响。方法应用序列特异性引物-聚合酶链反应技术,检测175例EV71脑炎病儿(感染组,包括轻症118例和重症57例)及221例健康体检儿童(对照组)IL-8-251位点单核苷酸多态性。结果 IL-8-251位点AA基因型在EV71脑炎重症组和轻症组的分布差异有统计学意义(χ2=8.975,P<0.05),且重症组等位基因A频率高于轻症组(χ2=7.845,P<0.01),但感染组基因型分布和等位基因频率与对照组比较差异无统计学意义(P>0.05)。不同基因型病儿年龄、发热天数、白细胞计数、C-反应蛋白水平相比较差异有显著意义(F=7.068、5.797,χ2=10.689、6.989,P<0.05)。结论 IL-8-251位点等位基因A携带者在感染EV71后易发展为重症脑炎。Objective To investigate the impact of interleukin-8(IL-8)gene polymorphism on risk of suffering from enterovirus 71(EV71)encephalitis and its pathogenetic condition. Methods Employing polymerase chain reaction with sequence specific primers(PCR-SSP)technique,the single nucleotide polymorphisms of IL-8in 175 patients with EV71encephalitis-including 118 mild cases and 57 severe cases-and 221 healthy control subjects was detected. Results AA genotype distribution of IL-8-251 between mild and severe EV71 encephalitis was significantly different(χ^2=8.975,P〈0.05),and allele A frequency in severe group was higher than that in mild group(χ^2=7.845,P〈0.01),but the difference of distribution of genotype and allele frequency between EV71-infected group and control group was not statistically significant(P〉0.05).The differences of age,days of fever,white blood cell count and level of C-reactive protein among sick children with different genotypes were significant(F=7.068,5.797;χ^2=10.689,6.989;P〈0.05). Conclusion When an allele A carrier of IL-18-251A/T infected with EV71 encephalitis that is easily developed to a severe one.
