摘 要:为研究番鸭细小病毒基因遗传变异特征,通过PCR技术获得了番鸭细小病毒结构蛋白基因VP3序列AH-MDPV-VP3,并将该序列与GenBank数据库中登录的9条番鸭、鹅和鸭细小病毒基因相应序列进行了比对分析。测序结果可见番鸭细小病毒VP3基因全长1 605bp,编码534个氨基酸。氨基酸序列同源性和进化分析表明AH-MDPV-VP3与国内两个番鸭细小病毒基因KC171936和KM093740同源性较高,分别为100%和98.3%,且三者之间亲缘关系较近,来源于共同祖先;而与鹅细小病毒,尤其与台湾分离的鸭细小病毒基因AY382891和AY382892同源性相对较低,分别为91.6%和90.8%,遗传距离也相对较远。同时可见番鸭、鹅和鸭细小病毒的VP3基因各自较为保守,仅有少数氨基酸发生变异。本研究初步获得番鸭细小病毒VP3基因分子特征,为下一步该基因在免疫诊断及免疫防治中的应用奠定了基础。To investigate variation status of muscovy duck parvovirus gene,the VP3 gene was cloned by PCR,which we named as AH-MDPV-VP3.And we compared AH-MDPV-VP3 with the sequence of nine VP3 genes of muscovy duck,goose and duck parvovirus in the GenBank database.By alignment of the ten gene sequences,the higher homologies between AH-MDPV-VP3 and the KC171936 and KM093740sequences of muscovy duck parvovirus isolated in China were found,100%and 98.3%repectively,indcating a close genetic relationship with a common ancestor of evolution.And the homologies with goose parvovirus were lower,particularly,the AY382891 and AY382892sequences of duck parvovirus isolated in Taiwan were 91.6% and 90.8%,the genetic distances also were farther.Moreover the VP3 gene of muscovy duck,goose and duck parvovirus were relatively conservative with several amino acid mutations.The molecular characteristics of muscovy duck parvovirus VP3 gene were preliminarily obtained.And this study laid the foundation of immune diagnosis and prevention and control of muscovy duck parvovirus.
