摘　　要：目的:探讨温度敏感性水凝胶Dex-PCL-HEMA/PNIPAAm对大鼠心肌缺血再灌注损伤的作用。方法:结扎SD大鼠冠状动脉左前降支30min后放松结扎线,制作心肌缺血再灌注(IR)模型,以仅穿线但不结扎冠状动脉者为模型对照组(Sham组)。将IR模型制作成功的大鼠随机分为2组:磷酸盐缓冲溶液组(PBS组):缺血周边区注射PBS;水凝胶组(Gel组):缺血周边区注射Dex-PCL-HEMA/PNIPAAm水凝胶。28d后行超声心动图、组织病理学及免疫组织化学检测。结果:与PBS组相比,Gel组左室舒张末内径(LVEDd)、左室收缩末内径(LVESd)减小,左室射血分数(LVEF)升高,心肌细胞凋亡指数降低,梗死减小,梗死区室壁厚度增加(P<0.05)。结论:心肌IR后缺血周边区注射Dex-PCL-HEMA/PNIPAAm水凝胶可抑制细胞凋亡、减少梗死面积、保护IR后大鼠心功能。Objective：To investigate whether the implantation of thermosensitive Dex-PCL-HEMA/PNIPAAm hydrogel can protect rat heart from ischemia-reperfusion injury.Methods：Rats were randomly divided into three groups,in which one group was subjected to sham surgery,the other two groups received ischemia/reperfusion（IR）surgery（30min coronary artery ligation followed by reperfusion）,with the injection of 100μl of the following into the perimeter of ischemic zone：phosphate buffered saline（PBS group）or Dex-PCL-HEMA/PNIPAAm hydrogel（Gel group）.Four weeks after the surgery,echocardiography,histopathology and immunohistochemistry were performed.Results：Compared with the rats in PBS group,the rats receiving Dex-PCL-HEMA/PNIPAAm hydrogel showed a significantly smaller left ventricle end diastolic diameter（LVEDd）,left ventricle end systolic diameter（LVESd）and higher levels of left ventricle ejection fraction（LVEF）（P〈0.05）.The infarction size and apoptosis index in Gel group were significantly lower than those in PBS group,and the infarction wall thickness of Gel group was significantly higher than that in PBS group（P〈0.05）.Conclusion：Thermosensitive Dex-PCL-HEMA/PNIPAAmhydrogel can protect rat hearts from ischemia-reperfusion injury by reducing infarction size and attenuating myocardial apoptosis and improve cardiac function.
