摘 要:在p H值为7.40的HMT-HCl缓冲溶液中,铕(Eu3+)能和依诺沙星(Enoxacin)形成络合物,发出Eu3+的特征荧光,最大激发和发射波长分别为330 nm和612 nm,狭缝均为5 nm,肝素的加入能使Eu3+的特征荧光显著增强。在线性范围0.117-5.85?g·m L-1内,肝素的浓度和荧光强度成很好的线性关系,线性方程为△F=-5.691+6.98 c(r=0.994)(c:μg·m L-1);检出限为0.06μg·m L-1。本方法用于实际样品的测定,灵敏度高、选择性好、干扰少、操作简单,结果令人满意。A new method for determination of Heparin by fluorometry method was established. In HMT-HCl buffer solution at p H 7.40, Eu3+ formed complex with enoxacin, which emitted characteristic fluorescence of Eu3+ with maximum excitation wavelength at 330 nm, maximum emission wavelength at 612 nm and slit width at 5 nm. Additional Heparin resulted in a remarkable increase in the Eu3+ characteristic fluorescence. Within the range of 0.117+5.85 μg·m L-1, the concentration of Heparin had a linear relationship with the fluorescent intensity, with linear equation of △F =-5.691 +6.98 c(r = 0.994)(c:μg·m L-1)and detection limit of 0.06 ?g·m L-1. This method generated high selectivity, high sensitivity and low disturbance,which had been applied to the determination of practical samples with satisfactory result.
