摘 要:目的探讨一种耗时少、检出率高的基因组DNA检测方法。方法采用改良的溴化乙锭染色法检测提取到的样本基因组DNA。采用分光光度仪进行敏感性验证,聚合酶链式反应确认该方法能否用来检测PCR产物。结果改良的溴化乙锭染色法显著减少基因组DNA的上样量,提高基因组DNA的检测敏感性,节省操作时间,无需loading buffer,并可粗略估计基因组DNA浓度。结论改良的溴化乙锭染色法只适用于初筛样本基因组DNA,不能用于评估DNA片段的长度。Objective To explore a genome DNA detection method with less time-consuming and high detection rate. Methods Genomic DNA samples were extracted and detected with improved ethidium bromide staining method. The sensitivity was verified with spectrophotometer meter. Polymerase chain reaction was used to confirm whether the method was available for detecting the PCR product. Results The improved ethidium bromide staining method significantly reduced the quantity of genomic DNA sample applied to gel, improved the detection sensitivity of genomic DNA and saved operating time with no need for loading buffer. In addition, it can roughly estimate the concentration of genomic DNA. Conclusion The improved ethidium bromide staining method can be applied to screening genomic DNA sample, but it cannot assess the length of DNA hag- meats.
