摘 要:目的:观察半夏泻心汤对糖尿病胃轻瘫(Diabetic gastroparesis,DGP)胰岛素抵抗中葡萄糖转运蛋白4(glucose transport4,GLUT4)和糖原合成酶激酶-3(glycogen synthetase kinase-3,GSK-3)表达的影响,分析其治疗糖尿病胃轻瘫的作用机制。方法:将120只大鼠随机分为空白组和造模组,造模组链脲佐菌素(sreptozotocin,STZ)腹腔注射后采用高脂高糖饲料连续喂养15天,选择大鼠空腹血糖(fasting plasma glucose,FGB)在11.1-30.0mmol/L的大鼠作为糖尿病胃轻瘫大鼠模型,按照血糖分层随机分为模型组、二甲双胍组(1.80g/kg)、半夏泻心汤组(10.20g/kg)、半夏泻心汤组(5.10g/kg)、半夏泻心汤组(2.55g/kg),再按被试因素施加方法连续灌胃7d。实验结束后禁食不禁水12h后取材检测。结果:与模型组相比,空白组、半夏泻心汤组(2.55-10.20g/kg)与二甲双胍组((1.80g/kg)实验组大鼠GLUT4表达明显增加;与模型组相比,半夏泻心汤组(5.10-10.20g/kg)与二甲双胍组(1.80g/kg)组大鼠GSK-3明显降低。结论:半夏泻心汤对糖尿病胃轻瘫大鼠胰岛素抵抗时有增加GLUT4的表达、降低GSK-3的表达的作用。Objective: To observe the influence of treatment with Banxiaxiexin decoction on glucose transport 4( GLUT4) and glycogen synthetase kinase-3( GSK-3) in insulin resistance in rats with diabetic gastroparesis and to analyze the mechanisms behind therapeutic effects of Banxiaxiexin decoction on diabetic gastroparesis. Methods: One hundred and twenty rats were randomly divided into blank group( n = 10) and model group( n = 110). The diabetic gastroparesis rats model of rats whose fasting plasma glucose( FGB) tendency in 11. 1 - 30. 0mmo / L feed by high fat and sugar for consecutive 15 days after intraperitoneal injection with sreptozotocin( STZ),randomly divided into model group,metformin group( 1. 80 g / kg),Banxiaxiexin decoction group( 10. 20 g / kg),Banxiaxiexin decoction group( 5. 10 g / kg),Banxiaxiexin decoction group( 2. 55 g / kg). Results: Compared to the model group,serum GLUT4 content in blank group,Banxiaxiexin decoction group( 2. 55 - 10. 20g/kg) and metfomin groups( 1. 80g/kg) significantly increased( P 0. 05). Compared to the model group,serum GSK-3 content in blank group,Banxiaxiexin decoction group( 5. 10 - 10. 20 g / kg) and metfomin group significantly decreased( P 0. 05). Conclusion: Treatment with Banxiaxiexin decoction increased serum levels of GLUT4 and decreased serum of GSK-3 in insulin resistance in rats with diabetic gastroparesis.
