摘 要:目的:探讨黄精多糖对STZ所致Ⅰ型糖尿病大鼠心肌炎症损伤的保护作用。方法:50只SD雄性大鼠随机分为空白对照组、模型对照组、黄精多糖250mg/kg、500mg/kg、1000mg/kg剂量组。后4组均使用小剂量链脲佐菌素STZ腹腔注射制备糖尿病模型,造模成功后,除空白对照组和模型对照组外,其余三组分别口服灌胃给予黄精多糖250mg/kg、500mg/kg、1000mg/kg连续灌胃给药12周后检测大鼠血糖以及血脂,用超声心动仪分别检测大鼠的左室收缩压(LVSP)、左室舒张末压(LVEDP)、左室内压最大上升速率(+dp/dtmax)、左室内压最大下降速率(-dp/dtmax);取左心室组织进行病理HE染色,观察心肌组织细胞结构改变;免疫组织化学和免疫印迹测定心肌组织中Toll样受体4(TLR4)、核转录因子(NF-κB)、巨噬细胞移动抑制因子(MIF)的表达。结果:与空白对照组比较,模型对照组大鼠血糖及血脂明显增高,免疫组化检测心肌组织中TLR4、NF-κB、MIF蛋白表达水平明显增高;与模型对照组比较,黄精多糖1000mg/kg血糖、甘油三酯(TG)、总胆固醇(TC)、低密度脂蛋白(LDL)明显降低,高密度脂蛋白(HDL)明显提高;左室舒张末压(LVEDP)以及左室开始收缩至左室内压上升速率峰值时间(t-dp/dtmax)明显降低,左心室收缩压(LVSP)明显升高,免疫组化检测心肌中TLR4、NF-κB、MIF蛋白表达明显减少。免疫印迹法结果显示,黄精多糖250mg/kg、500mg/kg、1000mg/kg与模型对照组比较TLR4、NF-κB、MIF蛋白表达下降,且黄精多糖1000mg/kg组变化比较明显。结论:黄精多糖对Ⅰ型糖尿病大鼠心肌有保护作用,其机制可能与降低血糖、血脂、抑制炎性反应有关。Objective : To explore protective effect of polygonatum sibiricum polysaccharide on myocardial inflammation damage in the rats with type Ⅰ diabetes and its mechanism. Methods: 50 SPF SD rats were randomly divided into five groups. They were normal group,model group and low( 250 mg / kg),moderate( 500 mg / kg) and high( 1000 mg / kg) dose Polygonatum sibiricum polysaccharide groups,ten rats in each group. Low dosage streptozocin( STZ,35 mg / kg) was used to induce type Ⅰdiabetes model of rats in the last four groups. After the success of modeling,except the normal group and model group,low,moderate and high dose of Polygonatum sibiricum polysaccharide at doses of 250 mg / kg,500 mg / kg and 1000 mg / kg was administered with stomach with Polygonatum sibiricum polysaccharide. After gavage for 12 weeks to detect rats blood sugar and fat. Using ultrasound cardiograph,detecting heart function of the rats include left ventricular systolic pressure( LVSP),left ventricular end-diastolic pressure( LVEDP) and left ventricular pressure maximum rising rate( + dp / dtmax),left ventricular pressure maximum decline rate(-dp / dtmax). Take the left ventricular for pathological HE staining,cell structure changes of myocardical tissue was observed. Immuno-histochemistry methods and western-blotting were used to determine levels of Toll like receptor 4( TLR4),nuclear factor-κB( NF-κB),macrophage migration inhibitory factor( MIF) in myocardial tissue of the rats. Results: Compared with the normal group,blood suger and blood fat increased significantly in diabetic rats( 24. 84 ± 0. 42 mmol / L,4. 20 ± 0. 06 mmol / L,3. 76 ± 0. 14 mmol /L,4. 32 ± 0. 16 mmol / L,0. 70 ± 0. 01 mmol / L).
