摘 要:目的:探究黄精多糖对STZ所致糖尿病大鼠肾脏的保护机制。方法:将大鼠随机分为5组:对照组、糖尿病组、黄精多糖1000、500、250 mg/kg组,后面4组采用小剂量的链脲佐菌素(Streptozocin,STZ)30 mg/kg尾静脉注射建立糖尿病模型,黄精多糖分别以1000、500、250 mg/kg灌胃12周,检测血糖、血肌酐及尿素氮;MASSON染色观察大鼠肾脏形态学变化;考马斯亮兰检测大鼠24小时蛋白尿;免疫组织化学、Western Blot方法测定肾脏组织转化生长因子β1(TGF-β1)、内皮素-1(ET-1)、型胶原(Col-I)蛋白表达。结果:与对照组比较,糖尿病组大鼠血糖及24小时尿蛋白明显增加,肾脏组织中TGF-β1、ET-1、Col-I蛋白表达明显升高;与糖尿病组比较,黄精多糖1000、500、250 mg/kg组血糖及24小时尿蛋白明显降低,肾脏组织中TGF-β1、ET-1、Col-I蛋白表达明显降低。结论:黄精多糖对糖尿病大鼠肾脏有保护作用,其机制可能与其降糖、抑制纤维化因子有关。Objective: To explore the protective effects of polygonatum sibiricum polysaccharide( PSP) on myocardial tissue in type diabetic rats induced by STZ. Methods: SD rats were randomly divided in to five groups as follows : the control group,diabetic model group,PSP1000,500,250 mg/kg groups. To establish the type diabetes model,small dose of streptozocin 30mg/kg in the tail veins was used. PSP1000,500,250 mg / kg groups was given ig for 12 weeks in the three PSP treatment groups. Fasting blood glucose serum creatinine( SCR) and blood urea nitrogen( BUN) was examined; MASSON staining observes morphological changes in the rat kidney; Bradford detects 24-hour urinary protein in rats. Protein expressions of nephropathy tissue TGF-β1,ET-1 and Col-I were also tested by immunohistochemistry and Western Blot method. Results: Compared with the control group,the levels of blood glucose and 24-hour urinary protein in diabetes model rats were increased significantly( P〈0. 05),TGF-β1,ET-1 and Col-I protein expression increased significantly( P〈0. 05). Compared with diabetes model group,the levels of blood glucose and 24 hours urinary protein of PSP1000,500,250 mg / kg groups reduced significantly( P〈0. 05),the protein expressions of TGF-β1,ET-1 and Col-I were also significantly reduced( P〈0. 05). Conclusion: The protective effects of PSP was demonstrated on nephropathy tissue in diabetic rats. Its mechanism is partly associated with lowering blood glucose,fibrosis factors involved and so on.
