川芎-天麻不同剂量配比对大鼠肝阳上亢型偏头痛的药效学差异研究

摘　　要：目的：比较川芎-天麻不同剂量配伍比例（4：1、1：1、1：4,川芎在前,以下均同）醇提物对肝阳上亢型偏头痛大鼠的药效学差异。方法：雄性大鼠随机分为空白对照组、模型对照组、阳性药西比灵组、川芎：天麻不同比例组,灌胃附子（2g/kg/d）和皮下注射硝酸甘油（10mg/kg）复制大鼠肝阳上亢证偏头痛模型,比色法测定大鼠血清一氧化氮合酶（NOS）活力与一氧化氮（NO）水平,Elisa法测定血清内皮素（ET）、血栓素B2（TXB2）、6-酮-前列腺素F1α（6-keto-PGF1α）、降钙素基因相关肽（CGRP）水平并计算NO/ET、TXB2/6-keto-PGF1α。结果：各比例组NOS活力、NO、TXB2水平、NO/ET及TXB2/6-keto-PGF1α比值均降低,ET含量均升高,4：1、1：4组CGRP水平降低,1：1、1：4组6-keto-PGF1α含量升高,与模型组比较,有显著性差异（P〈0.05-0.01）。不同比例组两两比较,1：4组降低血清CGRP水平、提高NOS活力、升高NO、6-keto-PGF1α含量的效果优于其它比例组,差异有显著性（P〈0.05-0.01）。结论：川芎-天麻不同比例配比醇提物对大鼠肝阳上亢型偏头痛药效学存在一定差异,综合评价,以川芎：天麻（1：4）组疗效较佳。Objective： To study the pharmacodynamic differences of extract with different proportion of Rhizoma Chuanxiong and Gastrodia Elata on migraine rat models with hyperactivity of liver-yang. Methods： Male SD rats were randomly divided into 6 groups： normal group,model group,Flunarizine group and different propotion of Rhizoma chuanxiong and Gastrodia Elata alcohol extract group. The models were induced by aconite alcohol extract（ ig 2g / kg / d,21d） and glyceryltrinitrate（ sc 10 mg / kg）. Then determined the content of CGRP,NOS,NO,ET,TXB2,6-keto-PGF1α in serum. Results： Serum NOS,NO,TXB2 and the ratio of NO / ET and TXB2/6-keto-PGF1α reduced while ET raised in each group compared with model group（ P〈0. 05 - 0. 01）,CGRP levels reduced in（ 4： 1,1： 4） groups and the level of 6-keto-PGF1αraised in（ 1： 1,1： 4） groups（ P〈0. 05 - 0. 01）. Among the 3 extract groups,（ 1： 4） group was more effective on reducing CGRP,NOS,NO,the ratio of TXB2/6-keto-PGF1α and raising 6-keto-PGF1α than other groups（ P〈0. 05 - 0. 01）. Conclusion： Different proportion of extracts were effective on the model of migraine with hyperactivity of liver-yang,and（ 1： 4） group had the best therapeutic effect.
