摘 要:目的:研究丹龙醒脑方对脑缺血再灌注损伤大鼠大脑海马区血管内皮生长因子(Vascular endothelial growth factor,VEGF)及其受体血管内皮生长因子受体(Fetal liver kinase-1,Flk-1)的影响。方法:用改良Longa栓线法复制大脑中动脉闭塞模型,2h后开放血流再灌注。大鼠造模成功后被分为6组;丹龙醒脑方14.76g/kg、7.38g/kg、3.69g/kg剂量组,尼莫地平10.8mg/kg组,模型组,假手术组,灌胃7天后处死大鼠取脑,采用免疫组化方法测定海马区内皮细胞计数及微血管密度、VEGF、Flk-1的表达。结果:模型组与假手术组相比,大鼠脑海马区内皮细胞计数及微血管密度、VEGF、Flk-1蛋白表达均增强;丹龙醒脑方14.76g/kg、7.38g/kg、3.69g/kg剂量组与模型组相比表达明显增强。结论:丹龙醒脑方可以促进脑缺血区VEGF及其受体Flk-1的表达,其抗脑缺血再灌注损伤的机制可能与增强血管新生因子的表达有关。Objective: To investigate Danlong-Xingnao formula effect on VEGF and Flk-1 of cerebral ischemia-reperfusion injury rats hippocampus.Methods: The MCAO rat model was created by modified Longa method,blood reperfusion opened after 2 hours. The rats were randomly divided into six groups( High-dose,moderate-dose and low-dose Danlong-Xingnao groups,nimodipine group,model group and Sham group).The rats were given by gavage once a day for 7 days. Microvessel density,VEGF and Flk-1 of rats hippocampus were detected by immunohistochemistry. Results: The microvessel density,VEGF and Flk-1 in model group were higher significantly than Sham group. Compared with model group and Sham group,groups by high-dose( 14. 76 g / kg),middle-dose( 7. 38 g / kg) and low-dose( 3. 69 g / kg) gavage increased significantly in expressions of microvessel density,VEGF and Flk-1. However,there was no statistical significance between Danlong-Xingnao groups and nimodipine group,and no statistical significance was found in the inter-groups of Danlong-Xingnao. Conclusions: Danlong-Xingnao could increase expressions of VEGF and Flk-1 of cerebral ischemia-reperfusion injury rats hippocampus. The mechanism was related to the increase of factors for angiogenesis.
