三草方水提取物联合拓扑替康抗小细胞肺癌的实验研究

摘　　要：目的:对三草方水提取物联合拓扑替康进行抗小肺癌肿瘤活性研究。方法:分别采用插块法和细胞接种法建立移植人小细胞肺癌H446的裸鼠模型,观察不同剂量及不同组合三草方水提取物联合拓扑替康抗小细胞肺癌肿瘤的效果。结果:三草方水提取物100mg/kg剂量组效果优于其他剂量三草方水提取物;在插块法建立肿瘤裸鼠皮下移植模型中,拓扑替康1.5mg/kg剂量组的T/C(%)为29.8%,三草方水提取物联合用药组的T/C(%)分别为20.1%和16.5%,拓扑替康组和联合用药组的肿瘤增长率、肿瘤体积与空白组比较有显著差异(P<0.01);在细胞接种法建立肿瘤裸鼠皮下移植模型中,拓扑替康1.5mg/kg剂量组的T/C(%)为30.5%,三草方水提取物不同剂量与拓扑替康联合用药组的T/C(%)分别为24.2%和15.2%,联合用药组的肿瘤增长率、肿瘤体积与对照比较有显著差异,表现显著的抑制肿瘤生长的作用。结论:三草方水提取物(100mg/kg)与拓扑替康(0.75mg/kg)联合,在抗小细胞肺癌动物模型中表现出一定的协同作用。Objective： The antitumor activity of sancao prescription（ 三草方） combined with topotecan on small lung cancer was studied. Methods： Establishment of nude mice model transplanted human small cell lung cancer H446 by inserted block method and cell inoculation method respectively to observe the effect of different dosage and different combinations of san cao prescription of aqueous extract of topotecan against small cell lung cancer. Results： The effects of san cao prescription water extract（ 100 mg / kg） were bette than san cao prescription water extract（ 200 mg / kg,50 mg / kg）. In the insert block method to establish tumor in nude mice model,topotecan dose of 1. 5mg / kg groups T / C（ %） was 29. 8%,san cao prescription water extract and topotetan of different dosage in combination group T/C（ %） were 20. 1% and 16.5%. There was significant difference between the topotecan group and the combination group tumor the growth rate of the tumor volume compared with the control group（ P 0. 01）,and was showed significantly inhibit the growth of tumor cell inoculation effect. In the establishment of tumor in nude mice model,topotecan dose group（ 1. 5mg / kg T / C%） was 30. 47%,san cao prescription water extract and topotecan of different dosage in combination group T / C（ %） were 24. 2% and 15. 2%,the growth rate of the combination group of tumor,tumor volume had a significant difference compared with the control（ P 〈0. 01）,showed significantly inhibit the growth of tumor effect. Conclusion： san cao prescription water extract（ 100 mg / kg） and topotecan（ 0. 75 mg / kg） were showed some synergistic effect in anti-small cell lung cancer animal model,and the effects were better.
