摘  要:目的:优化D-半乳糖致小鼠亚急性衰老实验中,腹腔注射法和皮下注射法的最佳造模剂量。方法:8周龄雄性ICR小鼠随机分为8组,分别为正常对照组、腹腔注射D-半乳糖组(1.25g/kg、2.5g/kg和5.0g/k)和腹腔注射D-半乳糖联合亚硝酸钠组(1.25g/kg+0.08g/kg-Na NO2),皮下注射组D-半乳糖组(0.125g/kg和0.5g/kg)和皮下注射组D-半乳糖联合亚硝酸钠组(0.125g/kg+0.08g/kg)。连续给药第7周测定各组小鼠空腹血糖情况,8周后解剖,计算脏器指数,检测血清、脑组织和肝脏组织中生化指标变化,同时对肝脏进行石蜡切片。结果:腹腔注射D-半乳糖2.5g/kg可显著提高血清中TC和TG含量,降低脑组织中GSH水平;皮下注射D-半乳糖0.5g/kg显著提高血清中TC、TG和脑组织中MDA含量,并降低脑中GSH水平;肝脏石蜡切片显示以上两组的小鼠肝组织排列疏松,体现衰老状态。结论:腹腔注射D-半乳糖2.5g/kg和皮下注射D-半乳糖0.5g/kg使小鼠整体表现出衰老症状,可分别作为D-半乳糖致小鼠亚急性衰老腹腔注射和皮下注射的最佳造模剂量。Objective: The aim of this paper was to optimize the dose of intraperitoneal( ip) injection and subcutaneous( sc) injection and compare the pros and cons in D-galactose induced mice subacute aging model. Method: Eight weeks old ICR♂ mice were divided into 8 groups,which were normal control group,G-galactose group intraperitoneal injection( 1. 25 g / kg,1. 25 g / kg + 0. 08 g / kg NaNO2,2. 5g/kg,5. 0g/kg) and subcutaneous injection group( 0. 125 g / kg,0. 125 g / kg + 0. 08 g / kg NaNO2,0. 5g/kg). All mice were subjected to inject for 8weeks,and the fast blood glucose( FBG) was measured in the first seven week. Brain,liver,kidney,spleen and thymus samples were removed and weighed to calculate the tissue indexes at the end of the experiment. Blood samples were collected to evaluate blood lipid and brain to detect the level of MDA and GSH. At the same time,liver was performed to paraffin slice. Result: Compared with the normal group,TC and TG in serum were significantly increased( ↑27. 79% 、↑169. 76%),and the activity of GSH was dereased( ↓26. 03%). The contentl and activity of TC,TG,MDA and GSH were also significantly changed( ↑9. 54%,↑61. 69%,↑49. 40%,↓15. 35%). In addition,the liver paraffin-slice of D-galactose groups( 2. 5g / kg; ip or 0. 5g / kg; sc) were in an aging state with liver tissue loosely arranged. Conclusion: Dose 2. 5 g / kg and 0. 5 g / kg were suggested as optimal dosage for ip and sc injection respectively for D-galactose induced mice subacute agingl.
