摘  要:目的研究r Lj-RGD3对凝血酶诱发小鼠肺栓塞死亡的保护作用,并比较等效剂量的肝素与r Lj-RGD3的出血倾向。方法以尾静脉注射凝血酶2 500 IU/kg攻击造成小鼠肺栓塞死亡,计算小鼠肺栓塞死亡比率和保护百分率以评价r Lj-RGD3对抗凝血酶诱发小鼠肺栓塞死亡的保护作用;小鼠给予等效(对凝血酶诱发小鼠肺栓塞死亡的保护作用)剂量的肝素及r Lj-RGD3后,测定尾出血时间(BT)。结果 r Lj-RGD3呈剂量依赖性降低凝血酶诱发小鼠肺栓塞死亡比率而升高保护百分率。等效剂量的r Lj-RGD3及肝素BT延长%分别为78.1%和323.8%,r Lj-RGD3是肝素的24.1%。结论 r Lj-RGD3具有保护凝血酶诱发小鼠肺栓塞死亡作用,且出血倾向小。Objective To investigate the protective effect of rLj-RGD3 on thrombin-induced pulmonary thromboem-bolism in mice and to compare the hemorrhage tendency of heparin and rLj-RGD3 . Methods Mice were challenged by i. v 2 500 IU/kg thrombin to induce mouse pulmonary thromboembolism. The mortality and protective percentage were recorded to evaluate the protective effect of rLj-RGD3 on thrombin-induced pulmonary thromboembolism. Mice were administrated by heparin and rLj-RGD3 at the same protective effect dose. Then the tail bleeding time ( BT) was recorded. Results rLj-RGD3 decreased the mortality and increased the protective percentage in a dose-dependent manner. rLj-RGD3 and heparin at equi-effective doses increased BT by 78. 1% and 323. 8% respectively. rLj-RGD3-caused prolongation of BT was 24. 1% as much as that of heparin. Conclusion rLj-RGD3 has a strong protective effect on thrombin-induced pulmonary thromboembolism in mice with minor bleeding tendency.
