摘 要:目的观察大黄素对高血压大鼠的降压效应及肾组织中基质金属蛋白酶2(MMP-2)和组织金属蛋白酶抑制剂2(TIMP-2)表达的影响。方法 48只SD大鼠随机分成正常组、假手术组、高血压组和大黄素组,每组12只,每周进行鼠尾测压;12周后收集肾组织样本,用荧光定量PCR及免疫组织化学染色观察MMP-2和TIMP-2表达。结果治疗8周后,大黄素组血压较高血压组明显下降(P<0.01),但仍比正常组和假手术组高(P<0.01)。高血压组大鼠MMP-2和TIMP-2表达较假手术组、正常组明显增加(P<0.01),而大黄素组则较高血压组明显降低(P<0.01)。结论大黄素能够降低高血压大鼠血压及肾组织中MMP-2、TIMP-2表达。Objective To investigate the effect of emodin on blood pressure(BP) and renal expression of matrix metalloproteinase-2(MMP-2) and tissue inhibitor of metalloproteinase-2(TIMP-2) in hypertensive rats. Methods Forty-eight SD rats were randomly divided into normal, sham, hypertension and emodin groups, each of 12 animals. The tail BP was determined weekly during the observation period. Renal expression of MMP-2 and TIMP-2 was detected by fluorescent quantitative PCR and immunohistochemistry after 12 weeks. Results BP in emodin group was lower than that in hypertension group but higher than that in normal and sham groups(P〈0.01). Expression of MMP-2 and TIMP-2 was increased in hypertension group compared with normal and sham groups(P〈0.01), whereas their expression was significantly lower in emodin group than that in hypertension group(P〈0.01). Conclusion Emodin can reduce BP and renal expression of MMP-2 and TIMP-2 in hypertensive rats.
