摘 要:目的利用荧光素钠作为造影剂,建立一种简便、直观的大鼠血栓动态观察方法。方法 66只SD大鼠,雌雄各半,随机分为11组。3组大鼠经尾静脉分别注射荧光素钠2、1、0.5 mg/kg,确定合适荧光剂量;2组大鼠经尾静脉注射合适荧光素钠后,颈总动脉、下腔静脉连续照射1 h,观察荧光激发有无血栓形成;其余6组大鼠,分别为35%Fe Cl3诱导颈总动脉血栓模型组、下腔静脉血栓模型组、相应肝素钠组(520 U/kg)和假手术组。于荧光体视显微镜下观察记录血栓形成过程,并与血管称重和组织病理学观察结果相比较。结果 1 mg/kg剂量荧光钠用于实验效果较好。荧光连续激发1 h均没有引起明显的动静脉血栓形成。血栓模型组血栓形成呈时间依赖关系,而相应肝素钠组血栓明显减少,与血管称重和组织切片观察结果一致。结论荧光素钠作为造影剂,可用于实验动物血栓形成动态观察,并具有安全、简便、直观的优点。Objective To establish a simple, intuitional method to observe dynamic thrombosis induced by fluorescein sodium in rats. Methods 66 SD rats, half male and half female, were randomly divided into eleven groups. 2 mg/kg, 1 mg/kg and 0.5 mg/kg of fluorescein sodium were intravenously injected into rats respectively to determine the right dose; Common carotid artery and inferior vena cava were illuminated 1h to evaluating thrombosis induced by fluorescein sodium of 1 mg/kg. Fe Cl3-induced common carotid artery thrombosis, inferior vena cava thrombosis in rats, and corresponding heparin sodium(520 U/kg) and sham operation group, were used for the continuous observation of thrombosis formation with fluorescence microscope, and comparison of thrombus wet weight and histopathological changes in the vessels. Results 1 mg/kg of fluorescein sodium was a better dosage for the observation of continuous thrombosis formation in rats. There was no thrombus in the vessels illuminated last for 1h. The thrombosis formation showed time manner dependent in both thrombosis model group, and thrombus was obviously reduced in corresponding heparin sodium group, which was consistent with the result of thrombus wet weight and histopathological change. Conclusion Fluorescein sodium as a contrast agent can be used for dynamic observation of thrombosis in laboratory animals, which is safe, simple and intuitional.
