摘  要:为进一步探明山羊酪氨酸相关蛋白酶1(TYRP1)基因的结构和功能信息,通过生物信息学方法对山羊TYRP1基因编码蛋白的一级结构、二级结构、亚细胞定位和三级结构进行预测和分析,同时构建TYRP1基因的系统发育树。结果表明:山羊TYRP1基因共编码537个氨基酸,其编码产物为不稳定的亲水蛋白。二级结构主要以α螺旋和β折叠为主,存在二硫键、信号肽和跨膜结构域。亚细胞定位山羊TYRP1基因编码产物主要存在于内质网中;山羊TYRP1与绵羊的关系最为密切,其次是家牛、牦牛、野猪、羊驼、家猫、人和家兔,上述物种的系统进化情况与其同源性基本一致。说明,TYRP1基因编码产物在长期的生物进化过程中保守性较强。To further study on structure and function of C. hircus TYRP1 gene, the authors predicted and analyzed the protein primary structure, secondary structure, subcellular localization and tertiary structure of C. hircus TYRP 1 gene coding protein with the method of bioinformatics, and phylogenetic tree of TYRP1 gene was constructed as well. Results.. C. hircus TYRP1 gene encoded 537 amino acid, its coding product was unsteadily hydrophilic protein. The secondary structure was mainly composed of alpha helix and beta sheet, which had disulfide bond, signal peptide and transmembrane domains. Subcellular location of C. hircus TYRP1 gene coding protein primarily existed in the endoplasmic reticulum, which might play a role in the cell envelope, purines and pyrimidines, transport and binding, also involved in signal transducer,and effected the animal color. The tertiary structure of C. hircus TYRP1 gene was built by homology medeling. The phylogenetic tree of TYRP1 gene coding protein showed that C. hircus had the closet relationship with Ovis aries, followed by Bos taurus, Bos grunniens , Sus scro f a ,Vicugna pacos , Fells catus, Homo sapiens ,Oryctolagus cuniculus, the phylogenetic condition of above species with its homology were basically identical. It indicated that C. hircus TYRP1 gene coding product had a strong conservatism in the long-term evolution process.
