摘 要:目的:探讨健脾清化方对2型糖尿病大鼠血液基因转录水平表达调控的作用机制。方法:采用链脲菌素(STZ)诱导建立2型糖尿病大鼠模型,随机分为空白组、模型组和健脾清化方中药组,连续灌胃4周。抽提大鼠总核糖核酸(RNA),采用昂飞(Affymetrix)基因芯片杂交,进行聚类分析、基因功能(GO)分析和信号通路分析。结果:中药组基因谱与模型组的表达差异较大。中药组差异表达基因共392条。中药组对丝裂原活化蛋白激酶(MAPK)信号通路、酪氨酸激酶-转录因子(JAK-STAT)信号通路、血管内皮生长因子(VEGF)信号通路等的表达调控作用明显。结论:健脾清化方可能通过代谢通路中某些基因的表达来调控糖尿病大鼠血液转录水平,从而降低糖化血红蛋白,改善血糖水平。Objective: To study the mechanism of Jianpi Qinghua Decoction on blood gene expression profile in rats with type 2 diabetes mellitus(T2DM). Methods: The T2 DM rat model was established with streptozotocin(STZ) by intraperitoneal injection. 45 rats were randomly divided into the blank group, model group and Jianpi Qinghua Decoction group. All the groups were treated by gavage for 4 weeks. The Affymetrix gene chip hybrid technique was used to analyze cluster, gene function and signaling pathway of the total RNA extracted from rats blood. Results: Cluster analysis showed that there was a great difference in the genetic profile between the Jianpi Qinghua Decoction group and model group. There were 392 differentially expressed genes in the Jianpi Qinghua Decoction group compared with model group. Pathway analysis showed that Jianpi Qinghua Decoction could significantly regulate some signaling pathways, such as mitogen activated protein kinase(MAPK), tyrosine kinase-transcription factors(JAK-STAT), vascular endothelial growth factor(VEGF) and so on. Conclusion: Jianpi Qinghua Decoction might regulate blood transcriptional level in diabetic rats through the expression of some genes in the metabolic pathways to reduce glycated hemoglobin and improve blood sugar level.
