摘 要:目的:研究益气养血泄浊法对慢性肾功能衰竭(慢性肾衰)大鼠肾组织形态的影响及血管内皮生长因子(VEGF)表达的作用机制。方法:建立5/6(Ablation/Infarction,A/I)肾切除慢性肾衰大鼠模型,设假手术组、模型组、西药(氯沙坦钾+福辛普利)组、中药(肾衰Ⅱ号方)组,干预后检测肌酐(Scr)、尿素氮(BUN)、VEGF蛋白、VEGF m RNA,观察肾组织形态;结果:治疗后与假手术组比较,两治疗组Scr、BUN明显下降(P<0.05,P<0.01);肾衰Ⅱ号方上调VEGF蛋白及VEGF m RNA、降低Scr、BUN方面优于西药组(P<0.05,P<0.01);两治疗组病理变化改善明显。结论:肾衰Ⅱ号方可改善慢性肾衰大鼠肾功能,减轻肾纤维化,上调VEGF表达,其抗肾间质纤维化作用可能与调节肾组织VEGF表达信号转道通路,调节肾小管周围毛细血管生成效应,改善肾间质缺血缺氧相关。Objective: To study the influence of Yiqi Yangxue Xiezhuo therapy on renal histological of chronic renal failure rats and the mechanism of vascular endothelial growth factor(VEGF) expression. Methods: 5/6(Ablation/Infarction, A/I) nephrectomy-induced chronic renal failure rat models were established, and then the rat models were divided into sham operation group, model group, Losartan potassium+Monopril group and MRFD group. After intervention, serum creatinine(Scr), blood urea nitrogen(BUN) and m RNA and protein expression of VEGF were detected and renal histological was observed. Results: Compared with sham operation group, the levels of Scr and BUN in the two treatment groups were decreased significantly(P0.05, P0.01). The effects of MRFD on increasing the protein and m RNA expression of VEGF(P0.01) and decreasing the levels of Scr and Bun(P0.05) were superior to that of Losartan potassium+Monopril. The renal pathological changes of the two treatment groups were improved significantly. Conclusion: MRFD could improve renal functions, relieve renal fibrosis and up-regulate the m RNA and protein expressions of VEGF. The mechanism of resisting renal interstitial fibrosis possibly correlate with the regulating effects on VEGF signaling pathway of renal tissue and peritubular capillary angiogenesis and the improving effect on renal interstitial ischemic hypoxic.
