不同剂量葛根素对2型糖尿病患者血栓前状态分子标志物的影响分析

摘　　要：目的观察不同剂量葛根素对2型糖尿病患者血小板膜表面GMP-140(血小板α颗粒膜蛋白140)、血浆TXB2(血栓烷素B2)和6-酮-PGF1α(6-酮前列腺素F1α)及其比值的改变。方法选择住院2型糖尿病血栓前状态患者98例,随机分成A、B、C三组,分别给予葛根素200 mg、400 mg、600 mg静滴,每日1次,21 d为1个疗程。动态观察血小板膜表面GMP-140、血浆TXB2、6-酮-PGF1α及其比值,并与28名健康者的对照组比较。结果葛根素治疗后,各组GMP-140分子数均明显降低,均低于用药前和对照组(P均<0.05);各组血浆TXB2、6-酮-PGF1α及其比值,均低于用药前和对照组(P均<0.05),且随着葛根素剂量的增加,变化程度也增大。结论葛根素治疗2型糖尿病血栓前状态安全、有效,增加剂量,效果更明显。Objective To observe the influence of different dose of puerain on platelet surface factors such as GMP-140、plasma TXB2 and 6-keto-PGF1αand their ratio changing in Type 2 diabetes.Methods Type 2 diabetes mellitus in-patients with thrombus state before were assigned to A group,B group and C group. Three different groups were treated respectively with different dose of puerarin 200 mg,400 mg,600 mg with 0.9%NaCl 250 mL to peritoneal inject per day,21 days for one course of treatment. All patients were taken on an empty stomach blood at 6：30-7：30 before and after treatment. To be dynamic observing the GMP-140、plasma TXB2 and 6-keto -PGF1α and their ratio changing and comparing with related indicators in 28 healthy people. Results After treatment of peurain ,GMP-140’s number of molecules were decreased significantly（P 〈 0.05）,plasma TXB2 and 6-keto-PGF1α and their ratio were lower than before treatment and healthy patients（P 〈 0.05）,and with Dose increasing,the changing of the indexes were developing,Conclusion The influence of Puerarin on Type 2 diabetes mellitus in-patients with thrombus state before were safety and effectly,and the effect would be increasing with dose addition.
