摘 要:目的观察脑泰方对动脉粥样硬化兔氧化应激的影响,探讨脑泰方抗动脉粥样硬化的作用机制。方法采用"高脂饲料喂养+免疫刺激方法"建立动脉粥样硬化兔模型,模型成功后随机分为脑泰方组、模型组、阿托伐他汀组,另设空白对照组,每组10只。药物干预后,采用酶联免疫法测定兔血清SOD、MDA及ox-LDL含量;采用PCR及Western Blot技术检测LOX-1、VCAM-1、ICAM-1及MCP-1相关基因及蛋白表达情况。结果模型兔SOD较空白对照组显著降低(P<0.01),MDA及ox-LDL显著升高(P<0.01),主动脉LOX-1、VCAM-1、ICAM-1、MCP-1m RNA和蛋白表达较对照组显著升高(P<0.01)。药物干预后,与模型组比较,脑泰方组兔SOD明显升高(P<0.01),MDA及ox-LDL含量明显降低(P<0.01),LOX-1、VCAM-1、ICAM-1、MCP-1的m RNA和蛋白质表达明显下降(P<0.01)。结论脑泰方可能通过增强抗氧化能力,降低脂质过氧化程度及保护血管内皮的途径而发挥抗动脉粥样硬化作用。Objective To observe the effects of Naotaifang on oxidative stress in rabbits with atherosclerosis and investigate its molecular mechanisms. Methods The atherosclerosis rabbit models were prepared by feeding the high fat diet and immune stimulation. After the success of the model animals were randomly divided into Naotaifang group, model group,atorvastatin group and blank control group(n =10). After drug intervention,the serum superoxide dismutase(SOD)activity,malondialdehyde(MDA) and oxidized low density lipoprotein(ox-LDL) levels in rats were detected by enzyme linked immunos orbent assays. The LOX-1, VCAM-1, ICAM-1 and MCP-1 related gene and protein expression were detected by using PCR and Western Blot technology. Results Compared with the control group, serum SOD activity in tat models was significantly decreased(P〈0.01), serum MDA,ox-LDL was significantly higher(P〈0.01); m RNA and protein expression of aortic LOX-1,VCAM-1,ICAM-1,and MCP-1 were significantly increased(P〈0.01). After medicine intervention, compared with model group,SOD in rabbits of Naotaifang group was increased significantly(P〈0.01), MDA and ox-LDL were significantly lower(P〈0.01), m RNA and protein expression of aortic LOX-1,VCAM-1,ICAM-1,and MCP-1 were significantly lower(P〈0.01). Conclusion Naotaifang has anti-at he rosclerotic effects, it may through enhancing antioxidant capacity, reducing lipid peroxidation and protecting the vascular endothelium pathways.
