摘 要:研究不同精粗比全价颗粒饲料对犊牛不同组织中PepT1、PHT2和PHT1 mRNA表达的影响。试验选取日龄相近、体重相近的中国荷斯坦断奶公犊牛12头,分为4组,每组3头,各试验组精粗比分别为75:25(Ⅰ)、70:30(Ⅱ)、65:35(Ⅲ)和60:40(Ⅳ),预试期14 d,正试期56 d。结果表明:脾脏中PHT1mRNA表达量最高,肺中的PHT2mRNA表迭量最高,瘤胃中的PepT1mRNA表达量最高,而它们在心脏中表达量均最低;试验Ⅰ和Ⅱ组肝脏、皱胃、瓣胃和瘤胃中的PHT1 mRNA表达量极显著高于试验Ⅳ组(P<0.01),而试验Ⅲ组网胃的PHT1 mRNA表达量极显著高于其他各组(P<0.01);试验Ⅲ组犊牛肝脏和网胃以及试验Ⅱ组皱胃中PHT2 mRNA表达量极显著高于其他各组(P<0.01),试验Ⅰ和Ⅳ组瘤胃和瓣胃中PHT2 mRNA表达量极显著高于其他2组(P<0.01);试验Ⅳ组犊牛肝脏中Pep T1 mRNA表达量极显著高于其他各组(P<0.01),试验Ⅰ组瘤胃、网胃和瓣胃中PepT1 mRNA表达量极显著高于试验Ⅲ和Ⅳ组(P<0.01)。结果表明,不同精粗比全价颗粒饲料对不同组织的PepT1、PHT1和PHT2表达具有一定的影响作用,其中高精粗料促进其表达。To study the effects of concentrate to forage ratio in pellet feed on the expression of PHT1, PHT2 and PepT1 mRNA in different tissues. A total of 12 Holstein bull calf with the similar age and weight were divided into 4 groups with 3 calves. Calves in the groups were fed the pellet feed with concentrate: forage 75: 25(I), 70: 30(II), 65: 35(III) and 60:40 (IV), respectively. The preliminary trial period was 14 d and the experimental period was 56 d. The results showed that: the expression of PHT1mRNA in spleen, PHT2mRNA in lung and PepT1 mRNA in rumen were the highest, but the expression of them in heart were the lowest. The expression of PHTlmRNA in liver, abomasum, omasum and rumen of group I and II were significantly higher than that of group IV (P〈0.01), the expression of PHT1mRNA in reticulum were significantly higher than that of other groups (P〈0.01). The expression of PHT2 mRNA in liver, reticulum of group III and in abomasum of group II were significantly higher than that of other groups (P〈0. 01). The expression of PHT2 mRNA in omasum and rumen of group I and IV were significantly higher than that of other groups (P〈0.01). The expression of PepTlmRNA in liver of group IV were significantly higher than that of other groups(P〈0.01) and that of in rumen, omasum and reticulum of group I were significantly higher than that of group III and IV(P〈0.01). In conclusion, the different concentrate to forage ratio in pellet feed could influence on the expression of PHT1, PHT2 and PepT1 in different tissues and high concentrate could improve their expression.
