摘 要:目的观察锰中毒对大鼠海马微血管的影响。方法成年雌性SD大鼠12只,随机分成锰中毒组(MIG)和对照组(CG),每组6只。腹腔注射氯化锰染毒3周,用过氧化物酶组织化学染色法染色,显示海马各区微血管。用Image-Pro Plus图像分析软件对海马微血管密度(microvessel density,MVD)、面积密度(microvessel area density,MVA)进行定量分析。结果 MIG大鼠海马CA1区MVD与CG比较有明显降低,差异有统计学意义(P<0.01);MIG大鼠海马CA1区、CA2区、CA3区和DG区MVA与CG比较明显升高,差异均有统计学意义(P<0.01或P<0.05)。结论锰中毒会导致海马区CA1区微血管数量减少,血管腔增大。提示,锰中毒对海马结构和功能的损伤可能与微血管数量减少有关。Objective To investigate the influence of manganese intoxication on hippocampus microvessel of rats. Methods Firstly, 12 female rats were randomly divided into two groups: manganese intoxication group(MIG)and control group(CG)with 6 rats in each group. The mouse model of manganese intoxication was established by intraperitoneal injection of manganese chloride for three weeks. Then we applied DAB straining method to make the microvessel in different parts of hippocampus appear. Image-Pro Plus was used to analyze the microvessel density (MVD)and microvessel area density (MVA)of hippocampus. Results Through comparing that in the CG, we found that MVD in CAlarea reduced significantly in the MIG (P〈0.01); while the MVA in CAlarea, CA2 area, CA3 area and DG area increased, significantly in the MIG(P^0.05 or P ~ 0.01). Conclusions Manganese intoxication may lead to an expansion of vessel lomen in CA1 of hippocampus and reducing the number of microvessel. The present study suggested that the structure and the function of hippocampus damaged by manganese intoxication may be related to the decrease of microvessel.
