反复力竭运动对巨核细胞分化的调控作用

摘  要:研究反复力竭游泳运动促进巨核细胞分化的作用机制。方法:30只3周龄健康雄性BALB/c小鼠随机分为安静对照组(CON)、反复力竭运动组(REE)。力竭运动组进行为期2周的负重力竭性游泳运动,并于末次运动后分别取血,进行血小板计数和westernblot检测单个核血细胞中PDGFR-α表达;使用过表达、敲低实验,检测PDGFR-α在K562细胞经PMA诱导向巨核系分化过程中的作用。结果:力竭运动后,PDGFR-α表达量下调、血小板计数增加;PDGFR-α抑制K562细胞经PMA诱导向巨核系分化过程。结论:力竭运动组通过抑制PDGFR-α表达而促进巨核系分化过程使血小板计数增加。The purpose of this study was to investigate the mechanism of repeated exhaustive swimming induced platelet increase. Thirty BALB/c mice were divided into control group(CON) and repeated exhaustive exercise group(REE). REE took exhaustive swimming exercise for 2 weeks, and then platelet count and PDGFR-α expression were detected. It was shown that repeated exhaustive exercise increased the platelet count and downregulated the PDGFR-expression. It could be concluded that repeated exhaustive exercise promoted megakaryocytic differentiation via regulating the PDGFR-α signaling pathway.
