高原制作改良罗氏培养基的条件探讨

摘　　要：目的利用蒸汽凝固器在高原制作改良罗氏培养基的温度和时间设定值,探索适合高原改良罗氏培养基制作的条件。方法利用标准配制改良罗氏培养基液,以推荐的85℃的温度、50 min的时间为基础,设置不同的温度、时间进行比对,同时通过无菌试验、H37RV标准结核菌株生长实验得出适合高原地区的温度、时间。结果标准配制的改良罗氏培养基液,蒸汽凝固器78℃、50 min为适合高原地区制作改良罗氏培养基的温度和时间。结论应用蒸汽凝固器时将温度设定为78℃,耗时50 min制作的改良罗氏培养基无菌试验合格、H37RV标准结核菌株生长旺盛,适合高原地区制作改良罗氏培养基,有推广应用价值。Objective To improve the implementation of medium manufacture in steam coagulator in highland and explore the condition of Lowenstein- Jensen culture medium manufacture in highland. Methods Prepare the culture liquid according to the standard and treat the recommended 85℃,50 mins as bases and set- up different temperature time and compared,and the sterility test and H37 RV standard tuberculosis bacterial strain growth test were used to gained the proper temperature and time that are suited in highland in standard culture liquid. Results 78℃ and 50 mins are the proper temperature and time of the steam coagulator to prepare Lowenstein- Jensen culture medium in highland when standard culture liquid was used. Conclusions The Lowenstein- Jensen culture medium which was prepared in steam coagulator when the temperature was set at78℃ and took 50 mins passed through the sterility test and the H37 RV standard tuberculosis bacterial strain grew well,the temperature and time are proper to be implemented and worth popularization and application.
