摘 要:目的研究双合汤对酒精性股骨头坏死(AOFH)兔的血液流变学及血栓素A2(TXA2)、前列环素(PGI2)的影响。方法将60只中国白兔随机分成正常组、模型组、美多巴组以及双合汤低、中、高剂量组。除正常组外,各组给予中国白酒(乙醇含量56%)10 m L/(kg·d)灌胃。在造模的同时,双合汤低、中、高剂量组分别给予双合汤灌胃,每日早晚各1次,每次分别为50、100、150 m L。美多巴组给予美多巴灌胃,每日早晚各1次,每次0.125 g。正常组、模型组均给予生理盐水灌胃,每日早晚各1次,每次100 m L。兔给药8周后,取血测定兔血液流变学指标(全血高切黏度、全血低切黏度、血浆黏度、红细胞聚集指数、红细胞变形指数、卡松黏度、血沉方程K值、全血高切还原黏度、全血低切还原黏度、刚性指数)、血栓素B2(TXB2)、6-酮-前列腺素F1α(6-Keto-PGF1α)。结果与正常组比较,模型组中血液流变学指标、TXB2均显著升高(P<0.01),6-Keto-PGF1α显著下降(P<0.01)。与模型组比较,双合汤低剂量组与美多巴组可降低血液流变学指标、TXB2,升高6-Keto-PGF1α(P<0.05);而双合汤中、高剂量组可显著降低血液流变学指标、TXB2,升高6-Keto-PGF1α(P<0.01)。双合汤低剂量组中血液流变学指标、TXB2及6-Keto-PGF1α的水平与中剂量组比较差异均有统计学意义(P<0.05);双合汤低剂量组中血液流变学指标、TXB2及6-Keto-PGF1α的水平与双合汤高剂量组比较差异均有统计学意义(P<0.05);而血液流变学指标、TXB2及6-Keto-PGF1α在双合汤中、高剂量组中水平比较差异均无统计学意义(P>0.05)。结论双合汤能有效改善AOFH兔的血液流变学,降低血液黏度,纠正血中TXA2/PGI2失衡,从而防治AOFH。Objective To study the effect of Shuanghe Decoction on hemorheology, thromboxane A2(TXA2) and prostacyclin(PGI2) of alcohol-induced osteonecrosis of the femoral head(AOFH) in Chinese white rabbits. Methods Sixty Chinese white rabbits were randomly divided into normal group, model group, Madopar group and the low, medium,high-dose of Shuanghe Decoction group. Except the normal group, each group was lavaged with Chinese spirit(alcohol content 56%) 10 m L/(kg·d). Meanwhile, the low, medium and high-dose group were all lavaged with Shuanghe Decoction and the rabbits of them took Shuanghe Decoction two times a day with 50, 100, 150 m L each time. The Madopar group was lavaged with Madopar, and the rabbits took Madopar two times a day with 0.125 g. The normal group and the model group were given with physiological saline, and the rabbits of them took physiological saline two times a day,with 100 m L each time. After 8 weeks, rabbits were sacrificed to collect blood for determination of the indexes of hemorheology(high blood viscosity, low blood viscosity,plasma viscosity, red cell aggregation index, erythrocyte deformation index, ηc, K value of blood sedimentation equation, high blood reduced viscosity, low blood reduced viscosity, rigidity index), thromboxane B2(TXB2)and 6-keto-PGF1α(6-Keto-PGF1α). Results Compared with the normal group, the indexes of hemorheology and TXB2 in the model group were significantly increased(P〈0.01), and 6-Keto-PGF1αin the model group were significantly decreased(P〈0.01). Compared with the model group, the indexes of hemorheology and TXB2 were decreased in the lowdose of Shuanghe Decoction group and the Madopar group, and 6-Keto-PGF1αwere increased(P〈0.05). Compared with the model group, the indexes of hemorheology and TXB2 were significantly decreased in the medium and high-dose of Shuanghe Decoction group, and 6-Keto-PGF1αwere significantly increased(P〈0.01).
