摘 要:目的研究有氧运动对Ⅱ型糖尿病大鼠腹主动脉Apelin和一氧化氮合酶/一氧化氮(NOS/NO)系统的影响。方法雄性SD大鼠60只,随机抽取20只大鼠并分为正常对照组(C组)和正常运动组(Y组)。其余大鼠喂以高脂高糖饮食并腹腔小剂量注射链尿佐菌素的方法建立Ⅱ型糖尿病模型。建模后,随机抽取20只大鼠随机分为糖尿病对照组(DM组,n=10)和糖尿病运动锻炼组(DME组,n=10)。Y组和DME组大鼠进行无负重游泳运动(60 min/次,6次/周×6周),6周后检测空腹血糖、血清胰岛素、NO浓度、腹主动脉Apelin含量及NOS活性等指标;利用RT-PCR法检测腹主动脉中NO、Apelin、t NOS、e NOS和i NOS的mRNA表达。结果 (1)与C组相比,DM组大鼠空腹血糖水平(P<0.01)、血清胰岛素显著上升(P<0.01),胰岛素敏感指数显著下降(P<0.01);腹主动脉Apelin含量(P<0.01)、i NOS活性(P<0.01)明显增加,而总NOS(P<0.01)、e NOS活性(P<0.05)和血清NO含量(P<0.01)显著下降;Apelin(P<0.01)和i NOS(P<0.05)mRNA表达上调,t NOS(P<0.01)、e NOS(P<0.05)和NO(P<0.01)的mRNA表达下调。(2)与DM组相比,DME组大鼠空腹血糖(P<0.05)和血清胰岛素水平(P<0.01)显著降低,胰岛素敏感指数显著升高(P<0.05);血清NO含量(P<0.01),腹主动脉总NOS(P<0.01)和e NOS(P<0.05)活力显著上升,且i NOS活性(P<0.05)显著低于DM组;腹主动脉Apelin(P<0.05)含量显著下降;Apelin(P<0.05)mRNA表达下调,而t NOS(P<0.01)、e NOS(P<0.05)和NO(P<0.05)mRNA表达上调。结论Ⅱ型糖尿病大鼠腹主动脉内皮细胞生成NO的功能降低,从而反射性引起腹主动脉Apelin含量显著增多,出现"Apelin抵抗"。有氧运动锻炼可以显著改善Ⅱ型糖尿病大鼠腹主动脉生成NO的功能,降低Apelin含量,增强了Apelin/NOS/NO的舒血管效应。Objective The purpose of this study was to research the effects of aerobic exercise on Apelin concentration in aorta abdominalis and NO / NOS system of type diabetic rats. Methods 60 male SD rats,selected 8 randomly named as the C group( n = 8),the other rats fed with high glucose,fat diet and were injected STZ to set up type 2 diabetes mellitus models. After the model formating successfully. 20 type 2 diabetes mellitus rats were randomly divided into two groups,named as DM group( n = 10) and DME group( n = 10). The DE group rats did a 60 minutes’ unload swimming every day,and 6 weeks’ later,fast blood glucose correlative indexes and Apelin content in aorta abdominalis were measured. The mRNA expression of Apelin,NO,t NOS,e NOS and i NOS were all tested by RT- PCR method. Results( 1) Compared with C group,the fast blood glucose( P 〈 0. 01) and the insulin content( P 〈 0. 01) in DM group increased significantly,while the ISI( P 〈 0. 01)dropped sharply; besides that,the Apelin content( P 〈 0. 01) in aorta abdominalis and the activity of i NOS( P 〈 0. 01) rose obviously,while the activity of total NOS( P 〈 0. 01) and e NOS( P 〈 0. 05) in aorta abdominalis,and also the NO content( P 〈 0. 01)in serum dropped dramatically. The mRNA expression of Apelin( P 〈 0. 01) 、NO( P 〈 0. 01),t NOS( P 〈 0. 01) 、e NOS( P 〈 0. 05)and i NOS( P 〈 0. 05) all significantly changed( 2) Compared with DM group,the fast blood glucose( P 〈 0. 05) and the insulin content( P 〈 0. 01) in DME group decreased remarkably,while the ISI( P 〈 0. 01) increased apparently; meanwhile,the NO content( P 〈 0. 01) in serum,together with the activity of total NOS( P 〈 0. 01) and e NOS( P 〈 0. 05) in aorta abdominalis increased remarkably,furthermore,the activity of i NOS( P 〈 0. 05) decreased obviously compared with DM group,but the Apelin content( P 〈 0. 01) in aorta abdominalis dropped significantly.
