参七消痞丸有效成分提取工艺研究

摘　　要：目的筛选参七消痞丸中有效成分提取的最佳工艺条件。方法采用平行实验法以出油率为考察指标确定水蒸气蒸馏法提取挥发油的最佳时间。采用正交实验法以出膏率、柴胡总皂苷提取率及柴胡总皂苷含量为考察指标,选取加水倍量和煎煮时间为考察因素,筛选水提取最佳工艺条件。正交实验结果采用综合评分法进行统计分析。结果平行实验结果显示挥发油的最长提取时间为6h。正交实验结果显示第1次煎煮加水倍量、第2次煎煮时间、第2次煎煮加水倍量3个因素水平综合平均值的最大值分别为iii=0.738、ii=0.822、iii=0.807,极差分别为0.049、0.229、0.179。结论 6h可作为参七消痞丸挥发油提取较优工艺条件的一个参考值。其有效成分水提取工艺的较优因素水平搭配为第1次煎煮加水倍量为5倍、第2次煎煮时间为1.5h、第2次煎煮加水倍量为10倍。Objective To select optimum extraction process of effective component in ShenqiXiaopi Pills.Methods As examining index,oil extraction rate of volatile oil was used to determine the best time of steam distillation by method of parallel experiment.As examining index,ratio of extraction,radix bupleuri total saponins extraction yield and content of radix bupleuri total saponins were selected;as study factors,water addition and decoction time were selected to screen optimum process conditions of water extraction by orthogonal experimental method.A comprehensive scoring method was used to analyze results of orthogonal test.Results According to the results of parallel experiment,the longest time of volatile oil extraction was 6hours.According to the results of orthogonal test,maximum of integrated average of water addition of decoction for the first time,the second decoction time and water addition of decoction for the second time were iii=0.738,ii=0.822 and iii=0.807 respectively.The ranges were 0.049,0.229,and 0.179 respectively.Conclusion A period of 6hours can be used as reference of optimum extraction condition of volatile oil extraction in Shenqi Xiaopi pills.The optimum match of level of factors of effective component water extraction is 5times of water addition of decoction for the first time,1.5hours for the second decoction time,and 10 times of water addition of decoction for the second time.
