摘 要:目的 通过癃畅颗粒对大鼠前列腺增生组织凋亡调控基因fas表达的影响,探讨癃畅颗粒治疗前列腺增生的作用机制.方法 Wistar雄性大鼠120只,随机平均分为癃畅颗粒高、中、低剂量组,癃闭舒组,模型组和正常组6组,每组20只.除正常组外,其余各组采用大鼠去势后注射Testosterone Propionate(丙酸睾酮)至前列腺增生法造模,各组均灌胃给药30d后处死,摘取前列腺组织测量湿质量,计算前列腺指数,采用免疫组化PV法检测实验鼠前列腺增生组织Fas表达状况.结果 (1)前列腺湿重:正常组(0.62±0.91)g;模型组(0.92±0.50)g;癃闭舒组(0.75±0.58)g;癃畅颗粒低剂量组(0.80±0.92)g;癃畅颗粒中剂量组(0.79±0.10)g;癃畅颗粒高剂量组(0.64±0.71)g;与模型组比较,差异有统计学意义(P<0.05). (2)前列腺指数:正常组(0.142±0.061),模型组(0.225±0.064),癃闭舒组(0.168±0.092),癃畅颗粒低剂量组(0.199±0.047),癃畅颗粒中剂量组(0.182±0,036),癃畅颗粒高剂量组(0.167±0.039),与模型组比较,差异有统计学意义(P<0.05).(3)癃畅颗粒对良性前列腺增生大鼠前列腺上皮细胞fas基因表达影响(平均光密度):正常组(0.216±0.029),模型组(0,183±0.089),癃闭舒组(0.201±0.129),癃畅颗粒低剂量组(0.200士0.126),癃畅颗粒中剂量组(0.202±0.143),癃畅颗粒高剂量组(0.207±0.177),与模型组比较,差异有统计学意义(P<0.05).结论 癃畅颗粒能够上调大鼠前列腺组织fas表达,缩小模型大鼠前列腺湿质量,减轻病理变化,这可能是该药临床有效治疗前列腺增生症的作用机制之一.Objective To observe the effects of Longchang particles on apoptosis genes fas in prostatic hyperplasia tissues and explore its mechanism in the treatment of benign prostatic hyperplasia. Methods Total of 120 Wistar male rats were randomly divided into six groups including high-dose Longchang group, the medium-dose Longchang group, the low-dose Longchang group, Longbishu group, the model group and the normal group, and 20 rats in each group. In addition to the normal group, rats in the rest of the group were ovariectomized and then injected with Testosterone Propionate ( TP ) to establish the rat models with benign prostatic hyperplasia. Subsequence, the rats were intragastrically administrated with Longchang particles for 30 days. They were sacrificed and their prostate tissues were collected for wet weight measurement,expression ofFas gene. Results Prostate wet weight of the normal group was (0.62q-0.91) g, the model group (0.92q-0.50) g, Longbishu group (0.75±0.58) g, Longchang Granule of low -dose group (0.80±0.92) g, Longchang Granule of medium- dose group (0.79±0.1 0) g, Longchang Granule of high -dose group (0.64±0.71) g; with significant differences between the model and the intervention groups(P〈0.05).
