猫杯状病毒不同分离株免疫原性的比较

摘　　要：为确定猫杯状病毒（Feline calicivirus,FCV）分离株JL-1、JL-2、JL-3、JL-4、SH免疫原性强弱,将各株灭活后分别制备油乳剂疫苗。将6-12周龄健康非免疫家猫随机分为免疫组和对照组。免疫组皮下接种相应毒株制成的灭活苗,对照组接种等量F81细胞培养液。于接种后不同时间点采集猫血清,应用ELISA方法检测免疫猫Ig G抗体、IL-2、IL-4和IFN-γ含量,评价各株诱导的体液免疫水平;采用MTT法测定淋巴细胞增殖,评价各株诱导的细胞免疫水平。结果表明：各株均能诱导试验猫产生免疫应答,其诱导体液免疫顺序依次为JL-2〉JL-1〉JL-4〉JL-3〉SH,诱导细胞免疫顺序依次为JL-4〉JL-2〉JL-1〉SH〉JL-3,初步确定各分离株免疫原性顺序为JL-2〉JL-4〉JL-1〉JL-3〉SH。To identify immunogenicity of Feline calicivirus（ FCV）,five isolates were inactivated and then mixed with freund＇s adjuvant. Approximately 6-12 week-old healthy unvaccinated kittens were randomly allocated to vaccinated（ V） and control（ C） groups. Kittens of group V were vaccinated with five isolates respectively twice at a 2-week interval via the subcutaneous route,while group C received equal amount of F81 cell culture. The sera specific of all isolates were collected at different time. The content of Ig G,IL-2,IL-4 and IFN-γ was measured by ELISA Kit to evaluate humoral immunity level and peripheral lymphocyte proliferation measured by MTT assay to evaluate cell immunity level induced by five isolates. The results showed that all isolates could induce immune response of vaccinated kittens with the order of humoral immunity level： JL- 2〉JL- 1 〉JL- 4 〉JL- 3 SH,and cell immunity level： JL- 4〉 JL- 2〉 JL- 1〉 SH 〉JL- 3. These data demonstrate preliminarily that the immunogenicity order of five isolates was： JL- 2〉 JL- 4 〉JL- 1〉JL- 3〉 SH.
