摘  要:目的观察醒脑通络片(黄芪、川芎、桃仁、红花、鸡血藤等)对脑缺血损伤大鼠内源性神经干细胞的影响。方法将120只SD大鼠随机分为假手术组和模型组(生理盐水灌胃),醒脑通络片小、中、大剂量组5组,每组各24只,采用线栓法致大鼠脑缺血再灌注损伤模型。脑缺血再灌注后3、7、14、28 d时,神经功能评分后处死动物,用免疫组化法观察缺血侧海马溴脱氧尿嘧啶核苷(Brd U)、巢蛋白(Nestin)的表达。结果假手术组海马区仅有少量Brd U、Nestin阳性细胞表达,模型组及醒脑通络片各组再灌注3 d后Brd U、Nestin阳性细胞表达开始增多,7~14 d达高峰,28 d明显下降。和模型组比较,醒脑通络片各组Brd U、Nestin阳性细胞明显增多(P〈0.05或0.01)。醒脑通络片各组在再灌注14和28 d神经功能均显示恢复,和模型组比较有显著性意义(P〈0.05或0.01)。结论醒脑通络片可以促进内源性神经干细胞增殖、分化,可能为其治疗缺血性脑血管病的机制之一。AIM To observe the effect of Xingnao Tongluo Tablets( XNTLT)( Astragali Radix,Chuanxiong Rhizoma,Persicae Semen,Carthami Flos,Spatholobi Caulis,etc.) on endogenous neural stem cells in rats with cerebral ischemia injury. METHODS One hundred and twenty healthy SD rats were randomly and equally divided into the sham operation control group and model group( physiological saline both),low,medium and high-dose XNTLT groups. Middle cerebral artery occlusion model was made by string ligation. Brd U and Nestin positive cells were observed with immunohistochemistry after ischemia-reperfusion on the 3rd,7th,14 th and 28 th day,respectively,prior to the assessment of eurologic function score. RESULTS Only few of Brd U and Nestin positive cells were observed in hippocampus of the sham operation group. However,the Brd U and Nestin positive cells of other four groups began to increase on the 3rd day,peaks occurred from the 7th to the 14 th day and decreased obviously on the 28 th day after ischemia-reperfusion. Compared with the model group,the Brd U and Nestin positive cells significantly increased in the XNTLT groups( P〈0. 05 or 0. 01). The neurologic function recovered significantly in the XNTLT groups on the 14 th and 28 th day after ischemia-reperfusion with significant difference( P〈0. 05 or0. 01). CONCLUSION XNTLT promote the proliferation and differentiation of endogenous neural stem cells,which may be one of its mechanisms of action in treating cerebral ischemia.
